Anthocyanins as the water-soluble pigments widely exist in the purple crops. The most importantbiological activities of the anthocyanins are their antioxidant properties, free oxidative radical scavengingproperties and anti-cancer and so on. However, it is less reports that whether anthocyanins are endowedwith antiviral effects. Here, we attempted that purple crops anthocyanins had anti-acute viral TGEV andchronic oncovirus ALV-A in vitro, and explored the anthocyanins antiviral mechanism about ALV-Ainduced DF-1cell NFκB signal transduction pathway, it had important significance in poultry virusprevention and treatment.This study chooses purple corn bracts as raw material, through ethanol extraction obtainedanthocyanins extracts. Seven other kinds of anthocyanins extracts were choosed as research object, suchas purple cabbage, black soybean, blackcurrant, Sambucus williamsii hance, Vaccinium Uliginosum Linn,purple sweet potato and Raphanus sativus L.,their contents and structures were determinated by using pHdifferential and HPLC-MS methods. Based on cell model in vitro, TGEV and ALV-A were employed toinduce relevant ST and DF-1cell cytopathic effects respectively, and the inhibition effects of purplecrops anthocyanins were determined by MTT and microscope observation methods. Meanwhile,according to the theoretical basis about the NFκB signaling pathway predominate, purple cornanthocyaninss was choosed as an example to research that it dealed with ALV-A induced DF-1cells innucleic acid and protein levels, then the activity of cells NFκB p50/p65were determined by usingWestern Blot and EMSA methods. The conclusions are as following:(1) This study extracted11.54%anthocyanins from purple crop bracts, meanwhile the identifitionof the eight anthocyanins extracts showed that cyanidin is the basic unit of glycoside of purple corn,purple cabbage, black soybean, blackcurrant, Sambucus williamsii hance,Vaccinium Uliginosum Linn.,purple sweet potato, Raphanus sativus L.; while purple sweet potato was mainly peonidin, Raphanussativus L. was mainly pelargonidin.(2) During the safe concentration range, purple crops anthocyanins had importent anti-TGEVeffect and their sorts as follows: blackcurrant> black soybean> Sambucus williamsii hance> purplecorn=purple sweet potato> purple cabbage=Raphanus sativus L.> Vaccinium Uliginosum Linn.Monomer anthocyanins’ anti-TGEV effects were Cy-3-glc> Pg-3-glc. These datas could provide anobvious intuitive research model for anthocyanins antiviral effects.(3) During the safe concentration range, purple crops anthocyanins had importent anti-ALV-Aeffect and their sorts as follows: blackcurrant> black soybean=purple corn> purple sweet potato>Sambucus williamsii hance> Raphanus sativus L.> Vaccinium Uliginosum Linn> purple cabbage.Monomer anthocyanins’ anti-ALV-A effects were Pn-3-glc> Cy-3-glc> Pg-3-glc. These datas couldprovide a tumor model for anthocyanins antiviral effects.(4) On the tumor virus model, NFκB p50/p65activity were restrained that ALV-A induced DF-1cells dealed with by purple corn anthocyanins, it revealed the mechanism of anti-ALV-A from anthocyanins.
