Object:CMV has the broadest host range among plant viruses, and affect the agriculturalproduction seriously in Xinjiang. In this research, we use processing tomatoes, peppers andother vegetables materials to survey the CMV infection rates in Xinjiang, and also studied theCMV strain differentiation, restructuring, genomic variability, biological symptoms and CMVsatellite RNAs’ molecular variation in biology and genomic level in Xinjiang. And at last lay acertain foundation for preventive CMV in the future.Methods:DsRNA that extracted from pumpkin and processing tomato samples which collectedfrom Shihezi Vegetable Research Institute to CMV satellite RNAs and RNA3sequences cloneby RT-PCR; processing tomatoes, peppers, tomatoes, pumpkin samples which collected fromShihezi and Yining in Xinjiang was used to do serological research; purification CMV on C.Quinoa, and observe the bionomics on some plant;17RNA3related sequences was amplifiedby One-step RT-PCR, and4samples some genome sequences were amplified by Two-stepRT-PCR.Results:The length of The pumpkin satellite RNA N-sate and processing tomato satellite RNAsTo-10-sate, S9, S18-1and S18-2is335nt,381nt,334nt,396nt and334nt respectively. Thesequence comparison showed that they do not contain the yellow area which contained inCMVYSAT which cause yellowing symptoms on tomato and other host.The serological detection rate of the samples which collected from Yining and ShiheziVegetable Research Institute is that: CMV subgroup serotype: processing tomato74.15%(152/205), pepper8.6%(2/23), tomato75%(3/4), pumpkin100%(2/2); CMV subgroupⅡserotype were negative.The CMV sequences of RNA3of pumpkin samples N, YN-8, YN-9, tomato samplesYN-2, processing tomato samples To-10and S1-14was amplified; the sequences of CMV CPand part of MP of pepper samples YN-6, LJ-4, LJ-10and processing tomato samples S1-1,S1-3, S1-4, S1-18, S1-27, S2-76, S2-81, S3-13, S3-47, S3-59was amplified. Sequenceanalysis showed that all the sequences have the closest relationship with CMV subgroup B. And LJ-4and LJ-10sequence have low sequence similarity with other samples and that theyhave high variability than other samples, the CP sequences of them have closer relationshipwith U.S. tomato isolates X.RNA1, RNA2and RNA3sequences of NS0-4and LJ-12, RNA1, RNA2and part ofRNA3sequences of S2-58, RNA1and part of RNA2, RNA3sequences of M were amplifiedby RT-PCR. The complete genome sequence analysis showed that: LJ-12genome sequencehave high variation rate; sequences restructure phenomenon exsited in S2-58and LJ-12, andsequences of NS0-4and M does not presence.The symptoms observed results show that: the symptoms caused by NS0-4is later thanother three isolate, it need one month, but others need ten days. The symptoms the fourisolates caused is different: symptoms on Nicotiana glutinosa: NS0-4: Jue and brittle leaves;S2-58: petiole growth, leaves dark green and there was a group seat like leavescrisp; M:yellow, blade shrink curl, blisters, petiole variable length. Benthamiana symptoms: moreblades emerald green, blisters, shrinkage, branch; S2-58: leaves dark green, albino spot,slightly shrunken shape.Conclusions and presume:In this research we find that CMV subgroup B is widely popular in Xinjiang, and themolecular variance phenomenon is existed in them, especially the CMV which infect peppers;the sequences polymorphism of CMV and its satellite RNA is related with the region, but hasnothing to do with the type of host; the five satellite RNA do not contain the yellow areawhich CMVYSAT has.We conjectured that: the deletion of MP of NS0-4does not make the CMV loss theactivity completely, but only affect the movement of the virus; the recombination andmutation phi-phenomenon of CMV which infect pepper is necessary to complete theexpansion between peppers and pumpkin, processing tomato, Vigna umbellata and tomato,and the expansion between them is difficulter than the expansion among the pumpkin,processing tomato, Vigna umbellata and tomato.
