Dehydration affect plant gowth and development significantly, therefore, it is important to improve drought-resistant abality. Populus ussuriensis widely distribute in China, and play a important role in forestry research. In this study, we used DREB-transgenc plant and non-transgenic plant as object, and investigated the PEG6000-responsive transcriptome analysis of Populus ussuriensis via DGE method.We analyzed the dynamic transcriptome of DREB transgenic poplars under osmotic stress. Much of differentially expressed genes were shown, especially for up-regulated genes. After exposure to4%and7%PEG6000, respectively. Differential expression patterns throughout osmotic stress were identified for100genes. GO enrichment analysis showed that genes mapped to "small molecule metabolism" were significantly enriched. Gene expression analysis showed that nearly all of these genes were up-regulated. The differential gene expression profiles of DREB are useful for further analysis of arid tolerance mechanism underlying overexpressiong DREB gene.A total of1063genes were shown to be differentially expressed, and all these genes could be used as a late-stage study improve poplar drought-resistant ability of candidate genes.The mechanisms underlying dwarfism of were investigated using DGE method. According to the results a total of2612genes showed significant alterations in gene expression including1370up-regulated and1242down-regulated gnes. The gene expression profiles showed that DREB have a distinctive gene expression pattern. GO enrichment analysis showed that genes mapped to" photosynthesis" were significantly affected. Almost all of these genes were up-regulated. Our transcriptome analysis might provide some useful insights into the dwarf mutant formation.Due to have an excellent performance under arid stress, we analyzed the transcriptome of DREB under arid stress. Genes that only differentially expressed in were identitied. GO enrichment analysis showed that genes related to translation were significantly affected. Almost all of these genes were up-regulated. Our results might give some useful insights for further analysis of plant arid tolerance mechanism.
