Pleurotus sajor-caju as a fungi, which mycelia polysaccharides has many biological activity,such as antioxidant capacity, regulation of the immune,antitumor activity and so on.The purse of this study is to research the mycelia polysaccharides activities. Observe and record the influences of different microbial strain age of liquid fermentation; According to the fermentation broth pH, reducing sugar content and biomass, the growth line of Pleurotus sajor-caju mycelium was drew; research the mycelia polysaccharides antioxidant capacity in vitro. Single factor tests were done to analyze the influence on biomass and mycelia polysaccharides, including temperature、rotation speed、noculumsize、medium volume.Then the optimum the best suits culture conditions by using L34orthogonal design, extract mycelia polysaccharides. The experimental results show that:1. Pleurotus sajor-caju mycelia present logarithm growth in3~7d,9d reaches maximum value9.26mg/mL; as the growth of mycelium, reducing sugar consumption start falling rapidly, after the mycelium growth rate into the stationary phase and decline, reducing sugar content in about12mg/ml; the PH of fermented liquid maintained at5.5,2. The single factor tests demonstrated that the optimum fermentation conditions are as follows:temperature is28℃,rotation speed is120r/min, inoculum size is6%, medium volume is75mL/250mL. Orthogonal test results show that the suitable combination is A2B1C2D3of mycelium and mycelium polysaccharide, mycelium concentration of9.98mg/ml, mycelium polysaccharide content was4.95%.3. The best extraction yield of pleurotus sajor-caju mycelia polysaccharides is4.69%.4. The results of the vitro-anti-oxidation with the dosage of5.0mg/mL is the biggest activities,suggested that the clearance rate on hydroxyl free radical was58.9%; the clearancerate on superoxide anion was40%; the inhibition rate on auto-oxididation of erythrocytes was39.1%; the inhibition rate on liver cells Lipidperoxidation was 22.1%.5.The activities of SOD,T-AOC, GSH-px in serum of treatment groups are significant higher than control group (P<0.05).
