The symptoms of diseased rice, infected by Southern rice black-streaked dwarfvirus, were very similar to phosphorus deficiency symptoms, such as, diseased ricebecame dark blue, the heart leaf glowed slowly, leaves became rigor, dwarfism anddark blue, stripes and stunting became short necrotic, few panicles could be heading,whose head stage was later and smaller, seeds were fewer and seedling height waslighter. Inorganic phosphate concentration in rice was determined by the molybdenumblue method in the research, when healthy rice, in which SRBSDV could not bedetected, as the control. The result showed that the inorganic phosphate concentrationin healthy rice was3.23times higher than that in rice infected by SRBSDV. There wasa significantly difference between two groups (p<0.05), which proved that SRBSDVhad effect on inorganic phosphate concentration in rice.Using real-time PCR proved that OsPTF1gene and LOC_Os05g45350gene, thetarget gene of miR399, were differentially expressed (p<0.05) between health rice andrice infected by SRBSDV, with2.6and3.4times different, respectively, while otherphosphorus metabolism key genes, OsPHR1, OsSPX1, OsPSS1, OsTRXH1, were notdifferentially expressed (p<0.05). And miRNAs expressive quantity was determinedby miRNA chip technology, the results showed that the expressive quantity ofmiR399a, miR399d, miR399e, miR399h, miR399i, miR399j in health rice were2.67,2.60,3.37,4.08,2.70,2.81times, respectively, than those in rice infected by SRBSDV,all of them were significantly difference(p<0.05). It was found that phosphorusmetabolism probably was affected by the different expressive of OsPTF1gene andmiR399which were affected by SRBSDV.Using agrobacterium-mediated method to cultivate transgenic Nipponbare,miR399a over express transgenic Nipponbare plants were obtained. Positivetransgenic plants were detected by the PCR to amplify with resistance gene G418gene. Which could be the basis of the study on miR399function and the deeply study on the effect of southern rice black-streaked dwarf virus on phosphorus metabolismmechanism in rice.
