This study try to detection the purity of the self incompatibility groups by molecular markers. The main purpose of this study was to select Brassica napus precocious self-incompatible lines by Microspore culture and molecular markers. The double low self-incompatibility line which named S-32was used as female parent and three compatibility lines were used as male parent. The flower buds of F1was used for microspore culture. At the same time,30self-incompatibility lines were used to test the stability and feasibility of the molecular markers used in this study. The results are as followed:1. The molecular marker of S60-1+2and Spes1-5+6which were linked to self-incompatibility gene and self-compatibility gene respectively. These two molecular markers were used to test the thirty self-incompatibility lines including5600single plants. We also investigated the phenotype of this group. Maybe it’s the problem of DNA extraction or other reasons,12single plants has no band detected with the two molecular markers. The detection results showed that5520were homozygous self-incompatibility lines and68were homozygous self-compatibility lines. The result is consistent with the phenotype survey. Since these two molecular were stable and available.2. We get148regrowths by microspore culture at last and transplant them to the experimental field. There are61self-compatibility DH lines and18self-incompatibility lines according to the character in field. There are40haploid and29of the regrowths were died. In May2012,79DH lines were reproduced in LanZhou. For the reason of the late growth period,15DH lines didn’t get seeds. In this study we get64DH lines at last.3. Two self-incompatibility lines were selected in this experiment.11-9-5803-107was a good candidate material in consideration of the comprehensive characters. Who’s growth period was186days. The yield of one plant was higher than male parent, but lower than female parent. The content of erucic acid was less than one and glucosinolate was less than30umol per gram. The oil content was up to40.92percent, which was1.76percent higher than female parent and4.55percent higher than male parent. The growth period of self-incompatibility line of11-9-5803-112was higher than both of the parents, but it’s agronomic trait and quality chatacter were fine.
