The efficiency of Agrobacterium-mediated transformation of maize (Zea mays L.)immature embryos and the regeneration of transgeic callus are two crucial factorsrestricting the transgenic breeding efficiency. This study mainly explored the influence ofheat-shock pretreatment on Agrobacterium-mediated transformation efficiency of Hi-IImaize (Zea mays L.) immature embryos and the regeneration system establishment oftransgeic callus. The results showed as follows:(1) The pretreatment of immature embryos with heat-shock at38℃for9minutesand42℃heat-shock for3minutes could effectively improve Agrobacterium-mediatedtransformation efficiency. The new definition of effective accumulated temperature couldbe used to explain the phenomenon in this study.(2) The results show, high efficient medium of inducing somatic embryos is SⅠ+500mg/L CH+60-90g/L sucrose.(3) The results show, medium added0.5mg/L NAA can enhance adventitious rootformation; medium added2mg/L MET can promote the development of lateral roots,improve the length growth of all roots and strong seedling.(4)Each event’s seeds (T1) were harvested after self-intersection. One event with ahigher expression level of G6-EPSPS gene was found by molecular biological detection.
