Natural rubber is an important industrial raw material. Hevea brasiliensis, better known as the rubber tree, is the primary source of natural rubber, now it’s widely grown in tropical and subtropical as an important economic crop.3-Hydroxy-3-Methylglutaryl Coenzyme A Reductase(HMGR), Geranylgeranyl Diphos-phate Synthase(GGPS), vacuolar PPase(V-PPase) and Hevea rubber transferase(HRT) were crucial enzymes in regulating the biosynthesis of rubber. HMGR, GGPS, V-PPase gene and HRT (the gene family contains HRT1, HRT2, Hpt3, hcpt-3and HbCPT gene) genes have been cloned from rubber tree by predecessors at present, but their location on chromosome has not been reported.FISH and in situ PCR were used to physically locate the HRT, HMGR1, GGPS and V-PPase gene of Hevea brasiliensis and build its physical map in this research. Reveal the specific location of these genes, the distribution of characteristics and provide the basis of research for the rubber tree, genetic engineering, genetics and breeding. The results were as follows:(1) Chromosome specimens were prepared with immature leaves of Reyan7-33-97. The chromosomal locations of the HMGR1, GGPS, V-PPase, HRT1, HRT2and hcpt-3gene were determined by FISH detection:the HMGR1gene was located on the long arm of chromosome3, the percent distances from the gene locis to centromere was25.28;the GGPS gene was located on the short arm of chromosome4, the percent distances from the gene locis to centromere was72.39; the V-PPase gene was located on the short arm of chromosome10, the percent distances from the gene locis to centromere was25.78; the HRT2gene was located on the long arm of chromosome6, the percent distances from the gene locis to centromere was42.02; the HRT2gene was located on the short arm of chromosome1, the percent distances from the gene locis to centromere was53.84and the hcpt-3gene was located on the long arm of chromosome7, the percent distances from the gene locis to centromere was27.95;(2)FISH and in situ PCR were used to physically locate the Hpt3and HbCPT gene, and the same detection results have been got:the Hpt3was located on the long arm of chromosome7, the percent distances from the gene locis to centromere was25.23; the HbCPT gene was located on the long arm of chromosome9, the percent distances from the gene locis to centromere was50.75.
