A Study On The Impact Of Ovis Aries Offspring Gender With Testicular Injection RNAi Zfy Gene

Controling sex of livestock and producing livestock to meet the needs of graziery are always studied by animal husbandry researchers.Sex control of sheep is responsible for expanding the population and rapid propagation.The progress of spermatogenesis is controled and regulated by multifaceted environment factors in vivo.The study found that Zfy gene is required for Y spermatogenesis.RNAi is used for interferencing the expression of Zfy gene,and restricts or even damages the structure or function of Y sperm development,so as to achieve the purpose of sex control.Objective:In pape, in order to study the change of Ovis aries offspring gender ratio, the plasmid-mediated RNAi inhibits the expression of gender determination gene Zfy.Method:Constructing plasmid expression vector pSilencer/Zfy-shRNA, large quantity of recombination expression vectors were made and injected once per week into testis of male sheep and sustained3times.5days after the last injection, the semen was collected from ram for17days, simultaneously the superovulated ewes were conceived through artificial insemination and observed the sex ratio of offspring.Results:Born216lambs on gender statistics found that the34th day of testicular injection of carriers for breeding,the female lambs occupied81.8%, significantly higher than the normal sex ratio (P<0.05)Conclusion:RNAi recombination expression vector which inhibits the expression of sheep Zfy gene is constructed successfully,and the34th day of testicular injection of carriers for breeding can enhance the ratio of female sheep. Interfering the expression of Zfy gene that realized the sheep sex control before fertilization, and simultaneously lay a foundation for the study of other animals sex control before fertilization.
