Hongyang kiwi fruit, a kind of Hongxin kiwi fruit, is native to China. Because ofthe delicious taste and rich nutrients, Hongyang kiwi fruit have a high business value.However, Hongyang kiwi fruit is unable to preserve for over than one week, which leadto greatly lose of famers’ or businessmen’s benefits. Therefore, the long-termpreservation of Hongyang kiwi fruit has been an urgent problem needs to solve. In thisstudy, to solve the preservation problem and set up the platform for Actinidia genefunctional research, we developed Actinidia transgenic techniques to modify theHongyang kiwi fruit. In short, we firstly constructed a double-T vectorpCAMBIA1300-ACO-actin-term, the two T-DNA fragments in this vector were linkeddirectly. The drug-select gene hpt and target gene ACO was contained in two T-DNAregions respectively. The sequencing verified double-T vector with antisense ACO genewas then transformed to Agrobacterium tumefaciens EHA105strain. By using leaf dishtransformation method, the Agrobacterium tumefaciens EHA105contained thedouble-T vector was transformed to the Hongyang kiwi fruit. By PCR verification, wefound the trans-ACO gene has transformed into the regeneration plant successfully. Thisis the word first study by using the double-T vector to induce trans-ACO genecontaining vector to kiwi fruit and get the regeneration plant. Our study laid thefoundation for breeding of transgenic Hongxin kiwi fruit with long-term storability.Furthermore, to reduce the maturation period and increase disease-resistance propertiesof transgenic kiwi fruit, we grafted the transgenic Actinidia in vitro plants to theActinidia valvata, which was known as idea disease-resistant stock.In total,19of the20grafted transgenic plants were survived, indicating the survival rate was more than95%.The storability and disease-resistance properties of transgenic kiwi fruit will be testwhen we get the matured kiwi fruit in next year.
