The aim was to explore Fsp27gene expression rule of different pig breeds, the expression level of Fsp27in liver,subcutaneous adipose and longissimus dorsi tissues from Shanxi Mashen and Yarkshire pigs at different developmental phases was investigated and compared by quantitative real-time PCR and Western blot.①The real-time PCR results revealed that in Yarkshire pigs mRNA expresstion was relatively higher between2-3months, it was highest at4months of age and decreased at5and6months of age; mRNA expression of Fsp27in liver of Mashen pigs was highest at3months of age, then gradually declined rapidly; Fsp27mRNA expression of Yarkshire was higher than that of Mashen in different months. In adipose tissue Fsp27mRNA expression level of Yarkshire increased firstly and then declined, it was highest at6months; Fsp27mRNA expression of Mashen was highest at6months, which was significantly higher than other months; the expression level of Fsp27mRNA in Yarkshire pigs was high than that of Mashen at4months of age, but lower than that of Mashen in other months. Fsp27was poorly expressed in longissimus dorsi of Yarkshire Pigs in different months except the third month; in the fourth month Fsp27mRNA expression of Mashen pigs was highest than that in rest of months(P<0.01); the expression level of Fsp27mRNA in longissimus dorsi of Yarkshire was significantly lower that of Mashen pig at4months of age.②estern blot results showed that the protein expresstion in liver of Yarkshire pigs was higher between2-3months, the expresstion peak appeared at4-month-old of age, and then started decreased; the Fsp27protein expression of Mashen was highest at4months of age, then trended down between5-6months; Fsp27protein expression of Yarkshire pigs was higher than Mashen. In subcutaneous adipose tissue, on month5the Fsp27protein expresstion of Yarkshire pigs showed highest levels, which significantly higher than that of rest months; protein expresstion of Mashen pigs was higher at early developing stage, it was significantly lower on month5and was highest on month6; at2,3,5and6months of age Fsp27protein expression of Yarkshire pigs was lower than Mashen between breeds. In longissimus tissue,the protein expresstion of two breeds was higher almost every month; Yarkshire pigs protein expresstion was higher than that of Mashen pigs in different months except in April between breeds.③onclusion:the result suggested that the expresstion the role of Fsp27in different tissues are different between Yarkshire and Mashen pigs, which provid theoretical basis for further study the regulatory mechanism of lipid metabolism. The rule of protein expresstion was different from that of the mRNA,mRNA expression may be related to the post-translational regulation of the protein.
