[Objective] Effects of fluoride on B cell were studied by establishing fluorosis animal model, for further providing scientific basis for fluoride on humoral immunologic function.[Method] This study was composed of vivo experiments and vitro experiments. In vivo experiments, a mice fluorsis model was established by adding different concentration of sodium fluoride in drinking, infected7d,15d,30d,60d and90d, taken spleen, weighed and separated lymphocytes, with LPS stimulation, applied flow cytometry to detected the changes of expression of CDs, endocytosis on B cells, apoptosis rate and proliferation index in spleen cells. In vitro experiments, separated of spleen lymphocytes form mice, added LPS and different concentration of NaF into the culture medium. Applied flow cytometry to detected the changes of expression of CDs, endocytosis on B cells, apoptosis rate and proliferation index in spleen cells.[Results]1. In vivo results showed, the body weight gain and spleen index of mouse in every treatment groups were inhibited, the concentration of bone fluoride keep consistent relation with fluoride concentration, which was that the fluoried concentration rosed, concentration of bone fluoride increased significantly.. Compared with the control, the fluoride displayed significantly inhibition in the expression of surface marker, endocytosis on B cells and proliferation index in spleen cells, and the inhibition role showed dose-depended relationship. Different dosages of fluoride could lead to spleen cell apoptosis.2. In vitro indicated, compared with the control, different doses of fluorine gave rise to reduction of the expression level of CD86, CD40, CD138on B cells, apoptosis rate and proliferation index in spleen cells, but caused expression of CD80increased, When the fluorine concentration of50μmol/L, enhanced significantly of endocytosis, the rest of the concentration difference was not significant.[Conclusion] Excessive fluoride can lead to the changes of the expression level of CDs and endocytosis on B cells,, apoptosis rate and proliferation index in spleen cells, causing immunity dysfunction...
