Bemisia tabaci (Gennadius) and Trialeurodes vaporariorum (Westwood) are part of Homoptera,Aleyrodidae, both of them are worldwide pest of agriculture. In recent years as the pest number and areaincreased, the harm of crops and the surrounding environment has caused serious influence. As an invasivespecies, both at home and abroad of researchers has been keen to whiteflies thermal stability study. In viewof this situation, this experiment mainly proceed from Heat Shock Proteins, research its Heat shock factor1,while using Real-time PCR to observe the quantity control factor of expression under different temperaturecondition, and eventually to determines expression relationshipof hsps and hsf1.To further of stated themolecular mechanism of whiteflies outbreak. Its main conclusions are as follows:1. We obtain the part hsf1gene sequences (900bp) of Mediterranean(MED) and Trialeurodesvaporariorum by PCR technologies, and DNAMAN software analysis to verify that: the spread of genesequence conform to the hsf1gene sequence, hsf1conservative gene sequence similarity to92.30%, canbe used for quantitative validation tests.2. In view of the early stage of the laboratory, we cloned from hsp60gene full-length ofMediterranean (MED) and Trialeurodes vaporariorum using electronic cloning technology. Through NCBIonline ORF Finder features and combined DNAstar software analysis, we received full-length cDNA has1761bp open read box (open reading frame, ORF) and encoded587amino acids(aa), predicts its isoelectricpoint is9.18, protein molecular weight approximately49441Da. The gene5’UTR including143bp and3’UTR containing446bp in Mediterranean(MED). Hsp60of Trialeurodes vaporariorum full-length cDNAhas1725bp ORF, encoded575aa, predicts its isoelectric point5.3, protein molecular weight of about61133Da,the gene5’UTR including143bp and3’UTR containing the467bp.3. Using RT-PCR technique quantitative determination hsp60and hsf1in two whiteflies indifferent temperature,the results showed: two species gene are obviously cool gene expression, but there isno significant expression in the high temperature situation. And meanwhile we can see that hsf1expressionis positively correlated with the expression of hsp60, they were induced at low temperature at the sametime. This can be very clear description of hsp60gene induced by hsf1regulation factors and protection.
