Half-smooth tongue sole (Cynoglossus semilaevis) is an important aquaculture fish, Due tothe half-smooth tongue sole have high nutritional value. A large differences in male and femaleadult is that adult females are generally2-3times the adult males. And in the development ofprocess, Half-smooth tongue sole occur sex reversal under natural conditions. Therefore, in thestudy process, mechanisms for gender related decisions controlled studies should be part of a veryimportant. Based on the StAR gene cloning of Half-smooth tongue sole, The gene was found veryclear expression in the testis, suggesting that development of testes is closely related to the spermcells.It has done the foundation work and make pathe way to Half-smooth tongue sole breeding.Partial StAR gene sequence obtained from whole genome sequencing results of half-smoothtongue sole(Cynoglossus semilaevis) and its full-length cDNA sequence was acquired bySMART-RACE.The StAR complete cDNA was1294bp in length containing1470bp open readingframe (ORF) and encoded284amino acids,132bp5′un-translated region(5′-UTR) and310bp3′un-translated region(3′-UTR). Alignment analysis with other species and Homologousanalysis of the amino acids showed that StAR of Cynoglossus semilaevis was85℅similar tomost species of StAR including Solea senegalensis, Micropterus salmoides,Lateolabrax japonicusand Sparus aurata.RT-PCR results indicated that StAR mRNA was extremely higher expressed intestis, but was hardly expressed in the ovary of half-smooth tongue sole. From16days to66days,StAR gene is not Expressed in testis. While150days the expression was significantly increased,Until sexual maturity, StARmRNA had doubled the amount of expression. After the discharge ofsperm, the expression decline obviously.The result of in situ hybridization showed thatStARmRNA Expressed in Leydig cells of testis. Our results provide StAR gene was Sex-relatedgenes in half-smooth tongue sole,it played important role in the testis maturation and made greatfunction during the period of sperm formation.By gonads of Half-smooth tongue sole during different developmental stages forhistological analysis,We observed26days,46days having no significant differentiation ofgonads.When the86days,we observed Ovarian cavity apparent,this is a sign for females in gonadsto differentiation. Testis in the86d also generated a lot of secondary spermatocytes. This alsoshows that gonadal differentiation there is no clear distinction between defined gonadaldevelopment will be affected by many factors.By in situ hybridization, We completed a tissue localization about FTZ-F1gene and neurl3gene during the development of gonads.About FTZ-F1gene, we found Hybridization signal bothin females and males, In contrast above, testis have obviously strong hybridization signals. Testis of44days hybridization signals mainly in spermatogonia, female gonads of44days have a weakexpression in original primordial germ cells. The gene is mainly in the membrane of the oocyteexpression in an age of the ovary.About neurl3gene.The result showed that the gene hybridizationsignal only in the testis.In addition,Hybridization signals mainly in mature sperm cells. Weinferred from the above that neurl3gene made important role in the sperm cells occurrence anddevelopment of testis.
