The giant panda(Ailuropoda Melanoleuca) is unique rare and endangered species inour country, which has important scientific value, ornamental value and social value.Protection of the giant panda has becoming a common human cause, in which, the diseaseprevention and control of giant panda is an important part to protect the giant pandas.Many diseases, especially the viral infectious diseases have caused serious threat to thegiant panda. Studies have confirmed that giant pandas infected with CDV, RV(rotavirus),CCV, CAV and caused clinical signs.This is not only a serious threat to the giant panda population’s health and protection,and has the potential public hygiene problems, especially in captivity conditions. Now theepidemiological characteristics of viral infections, are not clear, lack of system research forcaptive giant panda, the main research still remain in the clinical diagnosis,epidemiological investigation, and a small amount of pathogen separation, etc., researchmethods is relatively backward, there is not a full investigation and study report for thegiant panda virus disease epidemiology up to now. So it is extremely urgent to establish thehigh-throughput, sensitive, rapid and accurate PCR or real-time PCR detection methods forsome important viral disease in captive giant pandas systemically.This study is mainly done the following work:1、Aiming at the giant panda may infectious viral diseases, including CDV, CPV, CCV,rabies virus(RV), CAV, CPIV, rotavirus(RV), the rapid, accurate and high specific PCRdetection methods were established respectively according to the conserved sequence.2、From46serum samples,30whole bloods,24cotton swabs and28feces samplestaken from Bifengxia Panda Base, Wolong Notional Natural Reserve, Songpan andYingjing area, the DNA or RNA were extracted. The molecular epidemiological survey formany important virus diseases in the giant panda was carried out by using the established PCR methods. The results is as follows: the regular PCR amplification from128samplesmentioned above for seven kinds of diseases are all negative.3、Taking the method of double antigen sandwich Elisa, the antibodies of46serumsamples against CDV, CPV, CCV, rabies virus (RV), CAV, CPIV and rotavirus (RV),were examined. The results showed that, there was one positive result for rotavirus (RV)antibody, and another one was positive for CDV, CCV, CAV, CPIV antibodiessimultaneously, the total positive rate was4.34%. All samples tested were negative forCPV antibodies. The serum samples were all taken from non-immune giant panda, andthese results indicated that the giant panda can infect with RV、CDV、CCV、CAV andCPIV.
