| Floral organ development-related genes not only determining the floral organ development, but also regulate the physiological and biochemical responses in plant. The study of floral organ development-related genes has an important significance to develop the molecular mechanism of floral organ development, and provides technology support is to molecular breeding. This paper adopted RACE technology to isolate6Floral organ development-related genes APETALA1(PIAP1), APETALA2(PIAP2), APETALA3-1(PIAP3-1), APETALA3-2(PIAP3-2), PISTILLATA (PIP1)å’ŒSEPALLATA3(PISEP3). On this basis, different expression patterns of these genes in different period and different organs were analyzed through real-time fluorescent quantitative technology to study the expression regulation mechanism of the genes in the process of herbaceous peony floral organ formation. A-class P. lactiflora genes PlAP1(GenBank accession number:KC354376) full-length is1106bp, the coding region is729bp; A-class P. lactiflora genes PIAP2(GenBank accession number:KC455454) full-length is1935bp. the coding region is1575bp; B-class P. lactiflora genes PIAP3-1(GenBank accession number:KC354377) full-length is895bp, the coding region is663bp; B-class P. lactiflora genes PlAP3-2(GenBank accession number:KC354378) full-length is844bp, the coding region is663bp; B-class P. lactiflora genes PlPI (GenBank accession number:KC354379) full-length is890bp, the coding region is627bp; E-class P. lactiflora genes PISEP3(GenBank accession number:KC354380) full-length is1165bp, the coding region is729bp. The homologous analysis showed that6genes share high homology with genes of other plant.2Gene expressions during floral development were also analyzed in different organs. The results show that the PlAP1gene was mainly expressed in the sepals, and the PIAP2gene was mainly expressed in the carpels and sepals. PIAP3-2and PlP1had the highest expression levels in the stamens, followed by the petals. The expression levels of the PIAP3-1gene (from highest to lowest) were in the following order:petals, stamens, carpels, and sepals. The PIAP3-1gene was mainly expressed in the petals and stamens, whereas the PlSEP3gene was mainly expressed in sepals and carpels.3In the petals with different degree of stamen petaloidy, the expression levels of PIAPI, PIAP2, and PISEP3gradually increases, whereas those of PlAP3-1, PIAP3-2, and PIPI gradually decreased. In herbaceous peony, the increased of A-Class gene and E-Class gene expression have a promoting effect to tamen petaloidy, the decreased of B-Class gene expression have a promoting effect to tamen petaloidy. |
