| Herein, Alcalase, As1.398, Neutrase, Papain, Protamex and Flavourzyme were selectedto hydrolyze meat of Rana sylvtica Le conte. According to the inhibition activity of oxidationthe proteases were chosen and got the optimum hydrolysis conditions. Under the optimumhydrolysis conditions, the optimum protease was chosen according to the antioxidationactivity of hydrolysate at the different time and the optimum condition for hydrolyze Ranasylvtica Le conte was determined. On the optimum hydrolysis condition of Alcalase, theantioxidation activity and the rates of hydrolyzation of Rana sylvtica Le conte were measuredat the different times. Then, compared the antioxidation with Vcã€BHTã€BHA at differentconcentration and had the animal experiment to determine antioxidant activity in vivo.The results showed that Alcalase was proved higher antioxidation activity and theoptimum hydrolysis condition for the meat of Rana sylvtica Le conte: T=55℃, pH6.5,E/[S]=4%,[S]=1.5%. The rate of hydrolyzation for the Alcalase was always rising, but theantioxidation activity of hydrolyzation was rising and then was to descend with the time. Theantioxidation of hydrolysate was the highest when DH was11.34%, and the ratio of inhibitionoxidation according to linoleic acid-KSCN method and pyrogallol method were28.4%,46.5%respectively. The ratio of antioxidation of hydrolysate from Rana sylvtica Le contehydrolyzed by Alcalase was1.58times,6.94times,20.62times as high as that of Vc with theconcentration of0.20‰,0.16‰and0.12‰respectively according to pyrogallol method. Butthe ratio of antioxidation was lower than that of BHT and BHA.The results of animal experiment indicated, the lowest dosage group in liver were notsignificantly enhanced(P>0.05)except the activity of CAT. But the activity of CAT, GSH-PXof the other groups were enhanced obviously(P<0.01)and the content of MDA of the othergroups in liver of mouse was significantly decreased(P<0.01)after feeding on thehydrolysate from rana protein hydrolyzed by Alcalase. So one can see the hydrolysate fromrana protein hydrolyzed by Alcalase can improve the ability of antioxidation obviously. |
