Florfenicol also known as Thi-amphenicol, as a derivative of thiamphenicol, is anew type of chloramphenicol broad-spectrum antibiotic, is currently the exclusiveantibiotic for animals, has good absorption, broad antibacterial spectrum, no residualin animal body or residues is low, no carcinogenic, teratogenic, mutagenic "threeletter" effect, and other has the advantages of no potential and anemia of the animal,has been widely used in veterinary clinic, it’s played a key role in prevention andtreatment of bacterial diseases of livestock and poultry. But florfenicol asThiamphenicol derivative, capable of generating reversible inhibition of red cell, havea certain effect on immune organs of the animal body immunity, bursa, thymus, cecaltonsils caused mild damage is irreversible in the short term, also can cause theimmune effect failure of vaccine. Immunity refers to the non-specific immunefunction, enhance the body immunity, enhance the body resistance material, alsoknown as immune adjuvant. In this study, SPF chicken, chicken as the research object,through the changes of cytokines in serum of the animal body to understand theimmune suppression, a comprehensive study of florfenicol inhibition of immune andinfluence on immune efficacy of Newcastle disease vaccine, the immune enhancingagent antagonistic florfenicol to florfenicol elimination effect, provide technicalsupport for clinical rational drug use and disease prevention and immunity. The mainresearch contents of this subject include the following three aspects:The first part:70chickens of10day old SPF were divided into seven groups (A,B1, B2, B3, C1, C2, C3group), A group was the blank control group, B1, B2, B3groups were in drinking water by adding water-soluble florfenicol100mg/L,200mg/L,400mg/L, C1, C2, C3groups were added florfenicol premix50mg/kg body weight,100mg/kg body weight, the body weight of200mg/kg in feed.11-17days of age ineach group of continuous administration of7d. In10,15,22,29,43,57days of agecollected serum for determination of IL-10and IFN-γ levels. The results showed thatthe water soluble florfenicol dose (200mg/L) and high dose (400mg/L), florfenicol premix dose (100mg/kg body weight) and high dose (200mg/kg body weight) showeda state of immune suppression, IL-10, IFN-γ content in serum increased.The second part: the effects of florfenicol on immune function of SPF ofNewcastle disease vaccine and used immunopotentiator antagonistic florfenicolinhibitory effect on SPF chicken immunity.50chickens of14day old SPF weredivided into five groups (A, B, C, D, E group). A group add in the feed florfenicolpremix200mg/kg weight; B group add water soluble florfenicol400mg/L; C groupadd in the feed florfenicol premix200mg/kg weight, add the Shen immune reinforcingagent1g/L22-28day age drinking; D group add water soluble florfenicol in drinkingwater400mg/L, add the Shen immune reinforcing agent1g/L22-28day age drinking;E group served as control.15-22days of age in each group of continuousadministration of7d, all groups at22days of age of Newcastle disease vaccineimmunity1plumes. In14,22,29,36,43,50,60days of age collected serum fordetermination of IL-10and IFN-γ and ND antibody level. The results showed that,after two formulations of florfenicol fed SPF chickens, both affect the immune effectof ND vaccine, the antibody titer levels lower than normal; Shen immunostimulantson SPF chicks in serum IL-10, IFN-γ secretion produced an inhibitory effect, cansignificantly reduce the serum IL-10, IFN-γ, eliminate the side effects of florfenic.The third part: the effects of florfenicol on Chicken Newcastle disease vaccineimmunity and application of immunopotentiator antagonistic florfenicol inhibitoryeffect on broiler immunity.50broilers of7day old broilers were divided into fivegroups (A, B, C, D, E). A group in the feed florfenicol premix200mg/kg weight; Bgroup add water soluble florfenicol400mg/L; C group in the feed florfenicol premix200mg/kg weight, add the Shen immune reinforcing agent1g/L18-24day agedrinking; D group add water soluble florfenicol in drinking water400mg/L, add theShen immune reinforcing agent1g/L18-24day age drinking; group E served ascontrol.11-18days of age in each group of continuous administration of7d,respectively in7,21day old immune Newcastle disease live vaccine1plumes. In7,10,18,24,31,38days of age collected serum for determination of IL-10and IFN-γand ND antibody level. The results showed that, after two formulations of florfenicolin feeding broilers, both affect the immune effect of ND vaccine, the antibody titerlevels lower than normal; Shen immunopotentiator to chicken serum IL-10, IFN-γsecretion produced an inhibitory effect, can significantly reduce the serum IL-10,IFN-γ, eliminate the side effects of florfenic.
