| Poplar is the high quality of industrial wood raw materials and good urban greening species.The main distribution region of south of poplar are Beijing, Tianjin, north China and so on. The transgenic research of poplar began in1986, more than20years poplar transgenic technology are developing rapidly. There are some important target genes, such as insect-resistant genes, resistance gene, salt gene, herbicide-resistant genes and reduce lignin gene has been transferred to the poplar.But the study of water resistance is not complete.It has important significance for afforestation and urban greening of Anhui by transforming the rice Sub1A-1submergence-tolerant gene-1to the poplar.In this study, rice cDNA were used as a template and cloned the Sub1A-1gene sequence by using RT-PCR from rice FR13A,subsequently to build the over-expression vector. Optimize the genetic transformation system in the poplar, and then transform the rice SublA-1gene over-expression vector into poplar. The results indicate that:1.We clone the flood-tolerant gene Sub1A-1from the rice variant line FR13A and the sequencing results show that the gene sequence is consistent with that NCBI published.2.The over-expression vector pBI121+Sub1A-1was successfully constructed by gene engeneering measure, such as digestionã€purificationã€ligation and transformation.3.Choose the orthogonal design, the poplar genetic transformation system had been developed, that is the genetic transformation effect is best when explants pre-cultured2d, the concentration of AS is100micro-mol/L, infected leaves time for15min, the bacterial concentration OD value is0.7and the co-cultured time is4d.4.Transform the rice SublA-1gene over-expression vector into poplar.Poplar strains which contain rice Sub1A-1gene was obtained after screening and molecular testing. |
