Dangshansu pear is native to Dangshan county of Anhui province. Because of bigfruit, pulp thick, crispy, juicy and sweet, it is not only the brand of fruit in China, but alsoone of the main export volume of varieties in our county. However, in resent years, as theresult of the variety degeneration and cultivation mismanagement and other reasons, thecontent of sclereid in Dangshansu pear increased and its pulp became thick and palatedreggy, which severely affected its taste and quality.Dangshansu pear is the typical self incompatibility in fruit trees. Pear has the obviousphenomenon of pollen xenia and pollen xenia may affect the quality of fruit. Therefore,screening appropriate pollinated varieties has great importance to improve the fruit qualityof Dangshansu pear. This experiment used different pollination varieties to pollinate withDangshansu pear and studied the influence of cross-pollination on the development ofstone cell and pear fruit quality. Using cDNA-SRAP technology to make differentialexpression of the genes analysis with the fruits of Dangshansu Pear which have beenpollinated with different pollination varieties, and to research whether pollination canchange Dangshansu Pear’s Maternal gene expression. It provided theoretical and practicalbasis for selecting excellent pollination varieties, and improving fruit Dangshansu pearfruit quality.The main research conclusions were as follows:1. In10pollination varieties on Dangshansu pear, the size and distribution density ofsclereid in Dangshansu pear fruit were bigger when chose Yali as the pollination variety.While the size and distribution density of stone cell in Dangshansu pear fruit were smallerwhen chose Matihuang and Huashan as the pollination varieties. The kernels ofDangshansu pear were bigger when pollinated with Shinseiki, Cuiyu, Cuiguan andHuashan. When chose Qiyuesu, Matihuang, Zisu and Yali as the pollination varietieskernels of fruits were smaller. The content of stone cell was higher near the core andmiddle flesh when Dangshansu pear pollinated with Cuiyu, Cuiguan and Matihuang.2. In10pollination varieties on Dangshansu pear, the stone cell content of Dangshansupear was lower with the selection of Cuiyu, Cuiguan, Matihuang, Zisu and Huashan aspollination trees. The stone cell content of pear were higher when chose Shinseiki, Yali andKosui as pollinizers. Selections of Yali, Shinseiki and Kosui as pollination trees, thesugar-acid ratios of Dangshansu pear were higher and it tasted sweet and had good flavor.The sugar-acid ratios of Dangshansu pear were lower and the quality of it was poorer whenchose Cuiyu, Cuiguan and Huashan as pollination trees. Therefore, we chose Shinseiki, Yali, Matihuang and Qiyuesu to be pollinated varieties in production.3. In10pollination varieties on Dangshansu pear, choosing Shinseiki, Cuiyu, Zisu, Yali,Matihuang and Kosui as pollinizers, the lignin content of Dangshansu pear was higher. Itwas the highest when chose Yali as pollinizer. While the content of lignin was lower whenDangshansu pear pollinated by Matihuang, Huashan Qiyuesu and Wonhwang. Hence, wechose Qiyuesu, Matihuang and Wonhwang to be pollinated varieties.4. In suitable pollination varieties of Dangshansu pear, the stone cell and lignin contentwere the highest when chose Yali as pollination cultivars while they were relatively lowerwhen chose Matihuang as pollination tree. Consequently, we used cDNA-SRAP analysison Dangshan Su pear which was pollinated by Yali and Matihuang. The results show that3924cDNA fragments are obtained by cDNA-SRAP technology with196-pair-primercombinations. Among of them, there were121cDNA differential fragments. At last,23differential fragments with excellent reproducibility were cloned and sequenced. Theresults of bioinfomatic analysis showed that the23differential fragments could beclassified into5functional families included transcription factor genes, unknown genes,genetically modified plants genome related genes, chromosome11and the genes related topollen germination and pollen tube elongation. The function of F-box protein andS-ribonuclease which were origined from the pollinizer of Yali were that they could inhibitthe pollen germination and pollen tube elongation. It was hypothesized that the genes areto be involved in the metabolism of lignin during the stone cell development of DangshanSu pear and pollination may affect the metabolic of lignin.
