Breeding and disease resistance are the most important parts of the animal husbandry.Some chicken, Shouguang chicken, Wshang Luhua chicken, JiBai chicken and SPF chicken,are the material basis for breeding and production. The chicken economic traits can beimproved by molecular techniques through detecting the candidate genes for egg productionperformance and disease resistance.It could supply molecular data to assisted selection and early selection fine local breedsto studiy on genetic diversity of Shouguang chicken by PCR-SSCP. The study worked ondisease resistance in Shouguang chicken, Wshang Luhua chicken, JiBai chicken and SPFchicken. Detecting the expression lecels of TNFSF13B and NRAMP1genes by qRT-PCRafter infection with the MDV.Single nucleotide polymorphisms of FSHβ gene of Shouguang chicken were studied.The result showed that there was one single nucleotide polymorphism in exon3(A2540C).Exon3of FSHβ was selected to screen the genomic variation by means of PCR-SSCP andsequencing in Shouguang chicken. The result showed that there were three genotypes (AA,BB and AB) in Shouguang chicken. Statistic analysis showed that age at first egg of AAgenotype of individuals was shorter than AB and BB genotype of individuals, egg number at43weeks of BB genotype of individuals were more than AB and AA genotype of individuals,abnormal egg number of AA genotype of individuals were fewer than AB genotype ofindividuals of Wenshang Luhua chicken, no significant differences were observed in age atfirst egg, egg number at43weeks and abnormal egg number among all genotypes ofWenshang Luhua chicken (p>0.05).The result showed that the expression of the TNFSF13B gene of Jinbai chicken in 7-day-trement was signifigantly higher than that in other chicken spieces (p<0.05). Theexpression of the NRAMP1gene of Wenshang Luhua chicken in7-day-trement wassignifigant higher than that of SPF chicken and Shouguang chicken (p<0.05). The effect ofMDV on other chicken trement was not signifigantly. Statistic analysis showed thatWenshang Luhua chicken have the highest disease resistance to MDV, SPF chicken have thelowest disease resistance to MDV.
