Eucommia ulmoides oliv. of the eucommiaceae is a very important tree species with high value in economy and medicine. Available resources of eucommia ulmoides mainly include of leaves of eucommia ulmoides, barks of eucommia ulmoides, samara of eucommia ulmoides and male flowers of eucommia ulmoides. Eucommia ulmoides contains abundant active ingredients such as chlorogenic acid, Aucuba, a-linolenic and gutta-percha. Studies show that effective components of eucommia ulmoides have effects of regulating blood pressure, anti-tumor, enhancing immunity, kidney-reinforcing, anti-oxidation, anti-aging, tocolysis, heparoprotective effect, cholagogue action and diuretic, and so on. These evidence show that eucommia ulmoides have great prospect of development and utilization which may be involved in drugs, food, feed, chemical and other field. According to content of effective ingredients, physical and chemical character of eucommia ulmoides resources, to develop the synthetic development and utilizationg of effective ingredients specifically will be the trend of promoting the utilization efficiency of eucommia ulmoides resources and improving the cost-effectiveness of related products.In order to establish a set of comprehensive extraction technologies for effective ingredients of leaves and samara of eucommia ulmoides, this paper build detection methods of aucubin and α-Linolenic acid of eucommia ulmoides by HPLC and establish extraction and separation technology for samara oil, gutta-percha, chlorogenic acid and aucubin in turn by supercritical CO2extraction and column chromatography technology. This extraction technologies could be used to efficiently and synthetically extract effective ingredients of leaves and samara of eucommia ulmoides with getting more for once, saving resources and reducing raw material costs. Main results are as follows:1. The optimal detection method of aucubin of eucommia ulmoides by HPLC was achieved on a C18(150mm×4.6mm,5mm) column, the mobile phase was1to10percent Acetonitrile, the flow rate was0.8ml/min, the detection time was30min, the column temperature was20℃and determination wavelength was set at208nm. The calibration curve was linear in the range of0.30-1.50ug, the average recovery for aucubin was84.3%, RSD was1.97%. And the optimal detection method of a-Linolenic acid of eucommia ulmoides by HPLC was achieved on a C18(150mm×4.6mm,5mm) column, the mobile phase A was acetic:water of0.1:1, the mobile phase B was80%Acetonitrile, the flow rate was1.2ml/min, the detection time was30min, the column temperature was35℃and determination wavelength was set at205nm. The calibration curve was linear in the range of1.67-8.31ug, the average recovery for α-Linolenic acid was111.11%, RSD was1.56%.2. This paper comprehensive extract the effective ingredients of leaves eucommia ulmoides by solvent extraction and colum chromatography, the results showed that the yield and purity rate of gutta-percha were2.34%and96.21%by synthrtic method, the yield and purity rate of chlorogenic acid were82.60%and45.10%by Column chromatography, and the yield and purity rate of aucubin were64.09%and27.82%by Column chromatography. Through the above technical parameters, it could get20.0-24.Og gutta-percha from1kilogram leaves of eucommia ulmoides, the purity rate was96.31%, it could get7.5-26.0g chlorogenic acid from1kilogram leaves of eucommia ulmoides, the purity rate was46.31%, it could get10.5-20.0g aucubin from1kilogram leaves of eucommia ulmoides, the purity rate was27.79%.3. This paper comprehensive extract the effective ingredients of samara eucommia ulmoides by supercritical CO2extraction,solvent extraction and colum chromatography, the results showed that the yield rate of samara oil was14.72%and the purity rate of α-Linolenic acid was63.21%by supercritical CO2, the yield and purity rate of gutta-percha were11.55%and97.43%by synthrtic method, and the yield and purity rate of aucubin were82.76%and57.06%by Column chromatography. Through the above technical parameters, it could get140.0-150.0g samara oil from1kilogram samara of eucommia ulmoides, the purity rate of α-Linolenic acid was63.19%, it could get110.0-116.0g gutta-percha from1kilogram samara of eucommia ulmoides, the purity rate was97.47%, it could get35.0-52.5g aucubin from1kilogram samara of eucommia ulmoides, the purity rate was57.11%.
