Rape is an important oil crop, rapeseed oil is the world’s second largest source of vegetable oil. Breed yellow-seed Brassica can not only improve the rapeseed oil content, but also improve the quality of rapeseed oil and feeding quality,that is an important research in today’s rapeseed breeding.This test is optimize the extraction process of proanthocyanidins which is in rapeseed coat. Admeasurement the oil content, water content, hull content, thousand seed weight in different colour seeds, and proanthocyanidin, lignin, flavonoids, water content in different colour seed coat.That is to analyze the relationship between the seed coat color and the content of these substances, and obtained the following results:1.The use of ultrasound-assisted method to optimize the extraction process of the the proanthocyanidins with acetone concentration of70%, solid-liquid ratio1:0.2(g L-1), ultrasonic treatment time of15min, extracted three times, in that condition the proanthocyanidins extraction reach the maximum amount.2.Different development stages of the seeds oil content and weight of Sichuan Yellow and black seed of near-isogenic lines seems similar trend, but the oil content, thousand seed weight, hull content, water content in different colors seeds reached a very significant difference. Lignin, proanthocyanidins content in the seed coat of black seed near-isogenic lines are higher than Sichuan yellow seed coat in each period.4. The total flavonoids content in seed coat of Sichuan Yellow and black seeds of near-isogenic lines are marked difference. Use LCMS-IT-TOF to qualitative detection flavonoid in yellow and black rapeseed coat extracting solution, found flavonoids content are different,5flavonoids (myricetin、theaflavin、Kaempferol-3-(p-coumaryl) glucoside、Quercetin3-O-methyl ether peracetate、Catechin/Epicatechin)exists only in black seed coat.
