Senecio faberii Hemsl is grown between690~2450m of compositae plants of the genus Senecio, endemic to China, is widely distributed in sichuan,yunnan, guizhou and other regions. Studies had shown that senecio had different toxicity in the different areas, and the previous studies were mostly focused on the chemical composition of the separation from senecio. In order to aquire the information from toxicity of Senecio faberii Hemsl,and to provide reference for the development and utilization of the Senecio faberii Hemsl from baoxin.The main study results are the following:1. The acute toxicity and accumulative toxicity of Senecio faberii Hemsl in miceThe acute toxicity and the accumulative toxicity tests of Senecio faberii Hemsl to mice were carried out by modified Bliss method and fixed dose method. The results showed that:the LD50of Senecio faberii Hemsl leaves in mice was62.282g/kg,with95%confidence interval was52.684g/kg~73.645g/kg; the LD50of Senecio faberii Hemsl in mice was81.653g/kg,with95%confidence interval was70.145g/kg~94.527g/kg, both of them belonged to low toxicity. accumulative coefficient K of leave and whole plant was more than5in acculumative test, respectively. The conclusions were that leaves and whole plant of Senecio faberii Hemsl had low acute toxicity and mild accumulation toxicity.2. The30-d feeding test of Senecio faberii Hemsl in miceSeventy mice were randomly divided into7groups:the groups of the whole plant were including high-dose group (5.444g/kg), mid-dose group (1.633g/kg), low-dose group (0.544g/kg);the groups of leaves were high-dose group (4.152g/kg),mid-dose group (1.246g/kg),low-dose group (0.415g/kg); control group was dosed with equivoluminal distilled water for30d. The general clinical manifestations, changes of body weight and food consumption were recorded during the experimental periods.Hematological analysis, organ index of heart, liver, spleen, lung, kidney, and pathological examinations were examed at the end of experiment.The results showed that:changes of body weight, routine blood and organ index of heart, liver, spleen, lung, kidneys in various dose groups had no statistical difference compared with control(P>0.05).AST, ALT values of mice in both of high-dose groups were higher than that of the control group(P<0.05).Histopathology examination showed that different degrees of damage were detected in organs of heart, liver, spleen, lung, kidneys in both of high-dose groups.3. The100-d feeding test of Senecio faberii Hemsl in ratsSeventy rats were randomly divided into six experiment groups and one control group. The rats were orally administrated with Senecio faberii Hemsl for100days, with the dose of the whole plant5.444g/kg,1.633mg/kg,0.544g/kg, the leaves4.152g/kg,1.246g/kg,0.415g/kg and distilled water respectively. The general observations, changes of body weight and food consumption were recorded in rats during the experimental periods.Hematological analysis, organ index of heart, liver, spleen, lung, kidney, pathological examinations and ultramicrostructure of liver were examed at the end of experiment.The results indicated that:organ index of heart, liver, spleen, lung, kidneys and routine blood in various dose groups had no statistical difference compared with control(P>0.05),The serum level of ALT was significantly decreased in4.152g/kg group compared with control(P<0.01).The levels of AST,ALP,T-Bili in leaves4.152g/kg group and ALT,ALP in5.444g/kg were significantly decreased(P<0.05).The body weight gains of both of high-dose groups rats were slow,food utilization ratios were reduced. Histopathological examinations indicated that different degree of lesions in the heart,lung, liver, spleen,kidney.The ultrastructure of liver observed that mitochondria and endoplasmic reticulum structural change.4. Special toxicity of Senecio faberii HemslSperm abnormality test, micronucleus test and reproductive toxicity were carried out to evaluate teratogenesis and mutation toxicity of whole plant and leaves, respectively. The results showed that sperm malformation rates in all of the high-dose, mid-dose groups were higer than control group (P<0.01), the main types of sperm malformation were tail folding, amorphous and no hook. The micronuclear rates was significantly increased in the both of high groups(P<0.01) and mid-dose group of leaves(P<0.05) compared with control.The final body weight and weight gain of pregnant mice, body weight of fetal were significantly higher in the leaves high-dose group than control(P<0.05). Rates of the absorptive fetus was increased compared with negative control group. The main teratogenic types were skeletal deformities in mice. Senecio faberii Hemsl had significant teratogenicity and mutation on mice.
