Abstracts:Lipopolysaccharide (LPS) is a component of the cell wall of Gram-negative bacteria, it can induce the same pathological response as Gram-negative infections can activate by TLRs mediating the host immune response. Host defense system in male reproductive organs plays essential roles in the maintenance of health of bird and fertility. The CD4+helper T cells stimulate macrophages and B cells, whereas CD8+killer T cells show cytotoxic effects on infected cells. Lipopolysaccharide (LPS) is a component of Gram negative bacteria. The aim of the study is to determine whether expression level of IL-8and T cells are recruited in the male reproductive organs in response to LPS.Matured roosters were i.v. injected with LPS (0.5mg/kg) or PBS (control). The testis, epididymis and ductus deferens were collected by and3or6h after injection, and then expression level of IL-8were measured with Real-time PCR. At the same time, cryostat sections of them were immunostained using antibodies to chicken CD4and CD8. The frequencies of immunopositive cells in the lamina propria and epithelium of rete testis, efferent duct and epididymal duct in the epididymis and ductus deferens were analyzed under a microscope with image analysis software.The results showed LPS significantly induced the expression of IL-8in testis, epididymis and ductus deferens of rooster. Namely, LPS significantly-induced the expression of IL-8in testis and epididymis after a3-hour induction, but LPS continued to induce the expression of IL-8in ductus deferens from3to6hours after LPS injection. The results of T-cell subtype localization showed a small number of CD4+and CD8+T cells were localized in the testicular interstitial tissues of LPS-injected and control birds. Both types of T cells were identified in the lamina propria and occasionally in the mucosal epithelium of the ducts in the epididymis and ductus deferens in all birds. The CD4+T cell frequency was greater in LPS group than control in the efferent duct but was not significantly different in the rete testis and epididymal duct between two groups, and their frequency in the ductus deferens was significantly greater in LPS group than control after6h of injection. The CD8+T cell frequency in the rete testis and region of interstitial cells, but not in the epididymal duct, as well as in the ductus deferens and efferent duct was significantly and extremely significantly greater in LPS group than control after6h of injection. These results show that both CD4+and CD8+T cells are recruited in response to LPS stimulation in male reproductive organs, suggesting that they may play roles in the local host defense of these organs.
