Corn processing by-products of corn bran and corn syrup used in this study,belongsto industrial waste. Saccharomyces boulardii,Bacillus licheniformis and Lactobacillusplantarum as probiotics. Preparation of a new compound feed probiotic product, the productusing vacuum packaging technology preservation, living bacteria number>1.0×109cfu/g.provide a theoretical basis for the preparation of composite probiotics, lay a foundation for thefuture industrialized production of the composite probiotics.In this thesis, composition of corn bran and corn syrup were analyzed, drying andweighing method for the determination of the amount of dry matter,3,5-dinitrosalicylic acidcolorimetry (DNS) to determine the content of total sugar and reducing sugar. kjeldahlnitrogen determination of the nitrogen content, determination of the corn syrup acidity andcontaining dissolved phosphorus content. The results showed that the experimental corn syrupfeed probiotic cultures provide nutritional elements such as carbon, nitrogen and inorganicsalts. The experimental corn bran not only used as a carrier, also provide some nutrition foranimals.Growth curve and generation determination of the Saccharomyces boulardii,Bacillus licheniformis and Lactobacillus plantarum. The acidic and Bile salts conditionstolerance of the three kinds of probiotic were determined by exposing washed cellsuspensions to acidic conditions (pH1.0,2.0,3.0,4.0and5.0) and different bile saltsconcentration (0%,0.05%,0.1%,0.2%,0.3%(W/V)of physiological saline for1、2、3and4hhours separately, then three potentially probiotic separately with two pathogens37℃staticco-culture4h, Plate colony counting method for the determination of the probiotic viablecells. Lactobacillus plantarum acid resistance is the most obvious, the Saccharomycesboulardii and Bacillus licheniformis also has good acid resistance. Bacillus licheniformis bilesalts tolerance is the most obvious, the Lactobacillus plantarum and Saccharomyces boulardiialso has good resistance to bile salts. These studies suggest that the three potentially feedprobiotic have certain acid and bile tolerance capacity, possibly have a prebiotic effect inpiglet role by antagonizing pathogens.The probiotic carbon source, nitrogen source, corn slurry concentration andphosphate were studied with single-factor and orthogonal test, elect the best conditions of theprobiotic culture, validation tests as the best medium combination. The number of livebacteria Saccharomyces boulardii can reach4.5×l08cfu/mL, increased by25%than that before optimization; The number of live bacteria Bacillus licheniformis can reach2.3×l09cfu/mL, increased by15%than that before optimization; The number of live bacteriaLactobacillus plantarum can reach8.4×l08cfu/mL, increased by40%than that beforeoptimization; The preparation of composite probiotics separately put in4℃refrigerator and25℃constant temperature of incubator to save, the number of viable probiotics wasmeasured every other month to calculation the survival rate, continuous determination of12months. Results are displayed in4℃refrigerated probiotics, preserved after6monthssurvival rate is still more than60%, and stored at25℃incubator of probiotics, the viablecount decreased rapidly.
