The laboratory has screened from Rex stool, which have probiotic properties such asprevent and treatt diarrhea and improve feed utilization, Bacillus probiotics Tu-1(Bacilluslincheniformis). Through the fermentation condition optimization, in the pilot of the Tu-1agents for high-living bacteria (spores) content, and to study the storage characteristics ofthe Tu-1microbial agent, and rex feeding tests show that the agents improve Rex gutmicroflora environment, improve the digestive enzyme activity and reduce fecal nutrientcontent, and provide a reference for further study of its probiotic properties and mechanism.The specific findings are as follows:1. Identified by single factor experiment and orthogonal experiment method theBacillus probiotics (Bacillus lincheniformis) Tu-1strains optimal medium and optimalculture conditions are corn flour1.0%, yeast extract1.5%, MnSO4·H2O0.07%、Na2HPO40.4%、NaH2PO40.2%, initial pH7.5, inoculum was4.0%, bottling volume of50ml (250mL flask), rotation speed200r·min-1in cultured at37℃for48h, optimization aftersporulation rate of95.03%, final biomass was5.15×109CFU/mL.2. Througu two tons pilot fermentation test fermentation broth of B.licheniformis Tu-1bacteria concentrations reach more than5.89×109CFU·mL-1; Bacillus rate of90%;fermentation broth by centrifugation, bacterial sludge viable cells>50×108CFU·mL-1,spray drying sinensis content of live bacteria (spores)>100×109CFU·g-1, and finally addaccessories can be formulated into a1.0×1010CFU·g-1of solid dosage forms.3. Agents storage test showed that Bacillus Tu-1agents moisture content should beless than10%, stored at room temperature, sealed, dark, dry place. After24months, thespore survival rate can reach85%.4. Rex feeding trial showed that the addition of0.1%the Tu-1agents in the feed canincrease the number of beneficial intestinal flora, reduce the number of harmful bacteria. Inthe Fecal the number of lactic acid bacteria, Bifidobacterium, Bacillus respectively have anincrease of27.36%,25.76%,26.12%compared with the control group; E.coli was reduced27.77%. In rex feces the average viability of neutral proteases, alpha-amylase, cellulaseand lipase of test group (average enzyme activity of the beginning of the test section7d to45d) than the control group increased by9.20%,18.52%,36.91%and32.18%. In rex fecescontent of crude protein, starch, crude fiber and crude fat content of the test group than the control group decreased by18.93%,38.72%,28.44%and26.50%, inorganic compositionand moisture content did not change much. The feed weight comparison of test group wasreduced16.18%than control group. Diarrhea rate of the test group than the control groupdecreased28.57%. Tu-1agents can regulate Rex gut microflora, prevent and treat diarrhea,improve feed conversion ratio.
