A cross between the hexaploid wheat Shandong066559having purple pericarp (Pp),purple anthers(Pan) and purple leaf sheath(Pls) and SN1391without anthocyaninpigmentation are performed to produce each hybrid generation (reciprocal cross F1, F2, F3andBC1F1backcross generation, BC1F2). Analysis of traits of three phenotype, the inheritancelaw are performed. By using molecular markers, the Pan and Pls genes are mappedPreliminary. The main results are as follows:1.The purple-grain color, behaving as maternal effect, is controlled by2pairs ofdominant and complementary genes, the ratio of9purple:7white in F3and1purple:3white inBC1F2is observed. For anther color, the ratio of1purple:3yellow in F2and1purple:1yellowin BC1F1is observed. purple anthers is controlled by a pair of recessive gene. It has beenobserved that purple leaf sheath occurring in wheat is inherited as monofactorial trait thesame as purple anther except it is controlled by dominant gene.2.The genes between controlling three characteristics are different. By chi-square test,the genes controlling purple grain color are genetically independent to that of purple anthercolor, but there are genetic linkage between the purple grain and the purple leaf sheath.3. The genes Pan and Pls are mapped in2A and7B chromosome. Through the methodof BSA to build relative characters DNA’s pools, using SSR primers to screen purple genes.By analysis using Joinmap4.0software, xgpw2206and xwmc170purple anther gene locationon the2A, genetic distance are7.1cM and2.8cM respectively; xwmc402and xgwm68purple leaf sheath gene location on the7B, genetic distance are4.9cM and4.4cMrespectively.
