Meat rabbit’s reproductive traits’ heritabilities are very low and conventional breedingtechniques are difficult to improve them. In order to improve these traits, this study aimed atfinding the SNPs in GnRHR, FSHβ and GDF9gene for molecular marker assisted selection.In this study,227meat rabbits including163come from Kangda strain2and64from Kangdastrain7, were selected and cultivated by Qingdao Kangda Ltd and Shandong AgriculturalUniversity to detect genetic variants by Mix-sequencing and PCR-SSCP. SAS and R softwarewere applied to analysis allelic frequency and relations between reproductive traits anddifferent genotypes. The main results are as follows:For GnRHR gene, two mutations were found in exon1and3---T229A and TG insertioninto13430bp. The result of χ2test showed that all the other population fitted the H-Wequilibrium situation (P>0.05). For T229A mutation on the first parity TNB and later parityNAB in strain2, the genotype BB was1.23and1.56more than AA as a significant effect (P <0.05). The mutation inserted into the base of TG in13430bp in strain7on21days’ weight,weaning weight, weaning number, first parity TNB has a significant effect (P <0.05). Thegenotype FF was higher than EF on21days’ weight, weaning weight, first parity TNB for80.10g,362g, and2.75respectively. The genotype EF was higher than FF on21days’number for1.88.For FSH β gene, two mutations were found in exon2---T695C and A919G. The result ofχ2test showed that all the other population fitted the H-W equilibrium situation (P>0.05). TheT695C mutation has a significant effect (P <0.05) in strain2on later parity TNB and NAB,as the genotype MM was higher than NN for1.75and1.81. While in strain7, the mutationhas a significant effect (P <0.05) on the weaning weight trait. The genotype MM was higherthan NN for161g. The A919G mutation has a significant effect (P <0.05) in strain2on bornweigh, first parity TNB, later parity TNB and NAB. On born weight and first parity TNB, CTwas higher than TT for81.75g and1.17. On later parity TNB and NAB, CT was higher thanCC for1.17and1.34.For GDF9gene, a gene mutation was found in exon2---T1650C. The result of χ2testshowed that all the other population fitted the H-W equilibrium situation (P>0.05). For the mutation on weaning weigh and number in strain7, GH was328g and3.31more than HH asa significant effect (P <0.05).GnRHR and FSHβgene haploid type and their interactions effect analysis found that MT,NC, NT for feeding rabbit21days number, first parity TNB and NAB, later parity TNB andNAB has a significant effect.The study of GnRHR, FSHβ and GDF9gene detected five mutations and analyzed theirrelationship with the meat rabbit’s reproductive traits. The results show that these three genescould be or cooperate with major genes to affect meat rabbit’s reproductive traits. Under thecertain conditions, these five mutations could be used in the meat rabbit selecting for highreproduction traits in molecular marker assisted selection programs to improve the level ofprolificacy.
