Cucumber green mottle mosaic virus (CGMMV), as a member of Quarantine Pests of Entry Plants, is mainly against watermelon, gourd, cucumber and other cucurbit crop. For Hainan Province, where is a unique place for breeding these plants, and the prevention and treatment to the diseases on the seeds are more urgent. In recent years, the suspected symptoms of plants infected with CGMMV have appeared more often in many places. In order to further confirm the conditions, causes and regular patterns of the occurrences, and to take measures to prevent the spread of the disease timely and effectively, by study six main species of cucurbit crops in Hainan from2011to2013, carried out a detailed survey and sampled for detection and identification through RT-PCR, the results were as follows:(1) Recognized the main features of the fruits, leaves and vines within the investigation covered the damage symptoms caused by CGMMV on the six species of cucurbit crops, such as watermelon, cucumber, melon, gourd, pumpkin and bitter gourd in the main cultivation area in Hainan and the situation of the seeds caught by CGMMV, then, enriched the preliminary evidence to diagnosing CGMMV in fields which was compared with results of the tests.(2) A pair of primers was designed according to the conserved region of coat protein gene sequence. Used watermelon leaf and seed infected with CGMMV as materials, and materials were amplified target band while the negative control were not amplified the target strip. And the sequence of the target band proved it was a fragment of the coat protein gene, and the results showed that using the seeds as material through RT-PCR method could detect CGMMV successfully.(3) Collected leaf samples and seed samples from11main areas, for example, Wenchang City and Sanya City. The results of RT-PCR showed that the vast majority of six species’samples such as watermelon, cucumber, melon, pumpkin, gourd and bitter gourd were negative.Only one watermelon sample from Sanya and one gourd samples from Wenchang were positive. It was made clear that CGMMV was only sporadically distributed in Hainan. (4) Survey and observation suggested that different regions of the seeds had different infected situations, and at the same area, different species of these crops did not infect with each other. These results indicated that the main reason for the CGMMV’s occurrence in Hainan was infected seeds.(5) Proposed the specific measures to control the spread of CGMMV according to results of the investigation and identification in Hainan.
