Late blight caused by Phytophthora infestans is a devastating potato disease. It is a characteristic of plant innate immunethat plants recognizepathogens and trigger defence responce. According to the different mechanism and substances of identification plant innate immunity can be divided into pathogen associated molecular patterns (PAMPs) triggeredimmunity (PTI) and Effector-triggered immunity (ETI). PTI as the first line of plant resistance to pathogen plays an important role in plant disease resistance reaction. Because of PTI to be indiscriminatly against invasion of all sortspathogensas broad-spectrum resistance, the study on the mechanism of PTI has important applications for potato breeding in disease resistance. NbSERK3was invovled in resistance toP. infestans in Nicotina benthamiana, the resistance toP. infestans decreased obviously after silenced NbSERK3. StSERK3A/BAK1was cloned from cultivated potato according to the sequence information of NbSERK3, and speculated it would have a certain influence to late blight resistanceof potato. The experiment aims to preliminary explore the founction of StSERK3A/BAK1in potato PTI pathwaythrough analyzing StSERK3A/BAK1gene expression pattern, disease resistance of transgenic strains and protein interaction. The main results are as follows:1. Biotic and abiotic stress induced gene results analysis show that expressionof StSERK3A/BAK1gene has not significantly go up or downafter treated with P. infestans, SA, MeJA, ETH and mechanical wound stress,indicating that the gene will not be subject to P.infestans, SA, MeJA, ETH and mechanical wound stress.It constitutively maintains a basic expression level in vivo.2. Overexpressing and silenced StSERK3A/BAK1transgenic potato andN. benthamiana lines were obtained.The late blight resistance assay showed that the resistance levelof overexpression lines have not been significantly improvedcompared with the control, while the resistance of silence strains decreased obviously. Silence lines showed biggerlesion area and fasterexpandition. Symptom is clear when potato plants wereinoculated with virus PVY0compared to overexpression lines and control. 3. qRT-PCR analysis expression of transgenic plants geneson SA and JA signal pathway showed gene’s expression levels were not significantly increase or decrease without inoculationP.infestans. Expression of MAPK and ROS related genes was higher in overexpression strains while lower in silence strains.4.Bimolecular fluorescence complementation technology shows evidence of StSERK3A/BAKl interacted with NbLRKl and horizontal resistance-related gene StLRPK1,which make it sure that NbLRK1and StLRPK1involved in P. infestans trigged PTI, Which lays foundation for the further investigation of the interaction between StSERK3A/BAK1and PRRs...
