Cucumber (Cucumis sativus L.) is an economically important crop and an ideal system for studying several important biological processes such as sex determination, gene transfer among nuclear and organellar genomes and phloem physiology. The parthenocarpy of cucumber is an important economic trait closely related to yield and quality and is the objective traits of protected cultivation.ARF genes play important roles in fruit development. One target of studies that Cloning and expression analysis of cucumber CsARF10genes subfamily tries to explain the role of cucumber CsARF10genes subfamily in cucumber growth, especially fruit development. The other one that proteomic analysis the effect of CPPU on cucumber tries to explain the physiological mechanism of cucumber fruit development.1. Cloning and Expression Analysis of Cucumber CsARF10Genes SubfamilyThree ARF10genes (Auxin-Respond-Factor) named CsARF10a, CsARF10b and CsARF10c, were screened from the cucumber genome database by protein homo logy BLAST and cloned from cultuar’8419s-1’by polymerase chain reaction (PCR).In present study, phylogenetic reconstruction indicated that the CsARF10a had close relation with SIARF10, while CsARF10b and CsARF10c had similarity with At ARF10. Promoter prediction showed that the promoters consisted of diversified multiple hormone response elements. The real-time quantitative PCR (RT-qPCR) analysis proved that the expression of cucumber ARF10genes could be regulated by different phytohormones or concentrations of1-naphthylacetic acid (NAA) and performed different expression patterns. It showed that CsARF10gene subfamily had high transcriptional levels either in roots,stems,leaves or in male flowers; In additional, the qPCR results showed that although the expression of CsARF10genes had similar transcriptional trend, the CsARF10a had high expression level while the mRNA of CsARF10b and CsARF10c were relatively low.2. Proteomic Analysis of Cucumber Fruit Development of CPPU on Non-parthenocarpic Cultivar’8419s-1’In order to study the Mechanism of fruit development, non-parthenocarpic cultivar ’8419s-1’was as material. The female flowers were treated by100mg-L-1CPPU. The GO analysis showed CPPU regulate the cucumber fruit development process involved in the stress response, embryonic biosynthesis and degradation, carbohydrate and energy metabolism, signal transduction and secondary metabolites processes. Filtered differential protein spots at1.5times up or down regulation, got78in all,23protein up-regulated,55proteins were down-regulated.
