Tetranychus cinnabarinus is one of the most important pest mites around the world. Resistance to different pesticides have been widely observed due to the small body size, fast reproduction speed, limited moving area, frequent access to pesticides as well as high inbreeding rate.miRNAs (microRNAs) is a type of non-coding RNA that regulates and controls, usually21nt, coded by the genome of eukaryotic organisms. They regulate the expression of genes by complimentarily pairing with the3’UTR of the target gene through the2-8nucleotides at the5’ side, and by degrading mRNA or inhibiting the translation of mRNA of the target gene. To figure out whether the regulation of miRNA plays a role in the resistance of Tetranychus cinnabarinus, we carried out the sequencing of small RNA of Tetranychus cinnabarinus with Illumina HiSeq2000and identified the miRNA of Tetranychus cinnabarinus. Later we studied their expression levels in the sensitive strain(SS) and Fenpropathrin resistant strain(FeR). The results are as follows:1. Through sequencing, we obtained24796487reads in the Fenpropathrin resistant strain (FeR),22001867reads in the sensitive strain. We found21232699sRNA in the FeR, while there’re7014169(33.56%) that matches with that in the genome of Tetranychus urticae. In the SS, there are19657179in which6578128(33.21%) matches that in the genome of Tetranychus urticae. They are mainly20nt (15.23%)、21nt (22.60%)、22nt (19.10%).23nt (10.20%)2. We did the alignment between the sRNA and the miRNA in the Tetranychus urticae miRBase20, in which75end products of miRNA and45precursors were identified and93new miRNA end products and64precursors were predicted. In total there are26202target genes predicted,55target genes that might be related to insects resistance. All there resistance-related genes might be regulated by39miRNA that’s already known and33new miRNA. Through miRNA bases editing analysis, only210977edited points were found in the SS, compared to390968in the FeR.3. Compare the miRNA expression between the SS and the FeR, we found12miRNA that expresses significantly different, among which4up-regulated and8down-regulated. We conducted verification of the12miRNA that expressed significantly different through qPCR, and there are10miRNA that demonstrated consistent expression trend with consistency83%.
