In recent years,Aeromonas veronii lead to aquatic animal infection and caused by a large number of deaths. At present,the prevention of the disease is main taken by chemical drugs, but the chemicals would not only cause damage to farming water and can remain in the body, affecting the quality of aquatic animals. At present, the prevention of bacterial disease by vaccine, not only environmental protection and very effective. Studies on Aeromonas veronii inactivated vaccine has not been reported in our country. Therefore, this experiment has carried on the following research on Aeromonas veronii and inactivated vaccine:1The study on Aeromonas veronii characteristics and inactivationAeromonas veronii as the research object, in4ways of artificial infection of Crucian carp by intramuscular injection, intraperitoneal injection, skin injury infection and immersion infection Intramuscular and intraperitoneal injection of the highest mortality rate, Were100%,80%, indicating that the best way of these two infections. Preparation of inactivated vaccine with the different concentration of formaldehyde at different temperatures, testing for sterility and safety. The results obtained:using a0.2%formaldehyde-inactivated at28℃condition in24h, not only the complete inactivation can be achieved but also protection immunogenicity for maximum. In acute toxicity tests on fish, the LD50of this Aeromonas veronii was1.6×107(cells). With4×108cfu/mL regression infection test, white snakehead mortality was100%, thereby indicating that this Aeromonas veronii has strong toxicity.2The study on Aeromonas veronii inactivated vaccine immune responses for Crucian carpThe prepared vaccine into whole bacteria group and adjuvant group to immunized in Crucian carp,secondary immunization and poison attack.Determination blood cell count, white blood cell classification percentage, phagocytic activity,serum antibody titers and RPS. The results showed that:little change in the number of red blood cells, white blood cell count is rising; monocyte percentage, neutrophil percentage, PP and PI increased and then decreased, Significantly higher than the control group (P<0.05); thrombocyte percentage have a certain decline, but did not change significantly, Significantly lower than the control group(P<0.05); lymphocyte percentage overall upward trend, compare with the control group with significant difference(P<0.05); The immune organ index has an upward trend, significantly higher than the control group(.P<0.05). The above results show that Aeromonas veronii inactivated vaccine has good immune protection of crucian carp.3The study on Aeromonas veronii inactivated vaccine immune responses for Grass carpThe prepared vaccine to immunized in Grass carp, divided into6groups, Secondary immunization and poison attack, Measure its SOD, CAT, AKP activity, white blood cell classification percentage, Immune organ index,serum antibody titers and RPS, the maximum of serum antibody tite on immunized group were1:320..1:640>1:640、1:1280, Significantly higher than the control group (P<0.05)the RPS were70.59%、76.47%、88.24%、88.24%, Significantly higher than the control group (P<0.05). Three kinds of enzyme activity were increased after immunized.In SOD activity, Adjuvant group2was significantly higher than the other three groups(P<0.05), The adjuvant group1was significantly higher than that of two whole bacteria group(P<0.05),the whole bacteria group2was significantly higher than the whole bacteria group(P<0.05).In CAT activity, The adjuvant group2was significantly higher than two whole bacteria group(P<0.05), There was no significant difference between the other immunized group(P>0.05). In AKP activity, The adjuvant group2was significantly higher than the other3groups(P<0.05), whole bacteria group2was significantly higher than the whole bacteria group1and adjuvant group1, between whole bacteria group1and adjuvant group1there was no significant difference(P>0.05) monocyte percentage and neutrophil percentage increased and then decreased, the monocyte percentage of adjuvant group2was significantly higher than the other groups(P<0.05), the neutrophil percentage of adjuvant group1was significantly higher than the other groups(P<0.05), between immunized group and control group has significant difference(P<0.05), thrombocyte percentage changed little, and the lymphocyte percentage was increasing, significantly higher than the control group. The immune organ index has an upward trend, significantly higher than the control group(P<0.05). The above results show that Aeromonas veronii inactivated vaccine could improve the immunogenicity and protective efficacy of Grass Carp, adding adjuvant can enhance the role of immunity.
