The experiments investigate the effect of adding crystalline or coated lysine in diets of Triploid crucian carp on growth performance, body composition, antioxidant ability, amino acid metabolism and serum free amino acids. To ascertain the coated lysine can promote the growth performance and which coated lysine adding in diets of Triploid crucian carp can improve growth performance best.The feeding trial has two parts:In Exp.1,The soluble loss rate of crystals and three kinds of coated lysine at different conditions. In Exp.2, The experiments investigate the effect of adding crystalline or coated lysine in diets on growth performance, body composition, antioxidant ability, amino acid metabolism and serum free amino acids of Triploid crucian carp.Experiment1:Assay the soluble loss rate of crystals and three kinds of coated lysine at different pH (pH=6.5,6.5,7.2,7.5,8.0), different time (t=0.5h,1h,3h), different temperature (20℃,28℃) with ninhydrin staining method. The results show that, With pH increasing, the soluble loss rate of different forms lysine were firstly increased and then decreased; When the pH=7.2, the soluble loss rate were maximum; With the temperature or time increasing, the soluble loss rate of different forms lysine were increased, Among them, the soluble loss rate of hydroxymethyl cellulose coated lysine were minimum.Experiment2:Five diets, including control group diets(containing33%protein), control group diet added with crystalline lysine, ethyl cellulose coated lysine, acrylic resin coated lysine and hydroxymethyl cellulose coated lysine were designed to feed Triploid crucian carp fingerling with initial body weight of (10.67±0.07) g for8weeks. After the test, Fasting one day, Then serum free amino acids were determined0.5h,1h,2h,3h and5h after intaking diets. The results show that:1. Growth performance:Compared with control group diet, there were no significant effect of adding crystalline lysine, ethyl cellulose coated lysine and acrylic resin coated lysine on weight gain(WG), specific growth rate(SGR) and feed coefficient rate (FCR)(P>0.05). There was significant effect of adding hydroxymethyl cellulose coated lysine on WG, SGR and FCR. WG was improved by4.93%(P<0.05)and FCR was decreased by3.87%(P<0.05).2. Physiological function:Compared with control group diet, total antioxidant capacity(T-AOC) and superoxide dismutase(SOD) were significantly decreased and malondialdehyde(MDA) was significantly increased of adding crystalline lysine, ethyl cellulose coated lysine, acrylic resin coated lysine and hydroxymethyl cellulose coated lysine(P<0.05). There was no significant effect of adding hydroxymethyl cellulose coated lysine on T-AOC, SOD and MDA(P>0.05). There was no significant effect of adding crystalline or coated lysine in diets on glutamate pyruvate transaminase(GPT)(P>0.05),but glutamic oxalacetic transaminase (GOT) was significantly increased(P<0.05).3. Serum free amino acids:Serum free amino acids absorbing peak values time are4.65h,1.95h,3.41h,4.64h and5.41h respectively of Triploid crucian carp after intaking adding crystalline lysine, ethyl cellulose coated lysine, acrylic resin coated lysine and hydroxymethyl cellulose coated lysine diets. And serum free amino acids:Serum free lysine absorbing peak values time are4.19h,1.99h,2.72h,4.18h and4.81h respectively. The results show that, serum free amino acids absorbing peak values were advanced by adding crystalline lysine and ethyl cellulose coated lysine compared with control group diet, were similar by adding hydroxymethyl cellulose coated lysine, were delayed by adding hydroxymethyl cellulose coated lysine.In conclusion, there was no promotion on growth of Triploid crucian carp intaking diets adding crystalline lysine, ethyl cellulose coated lysine and acrylic resin coated lysine. and antioxidant capacity of Triploid crucian carp was destroyed. The goal of Saving the cost by adding lysine could’t be achieve. The soluble loss rate of hydroxymethyl cellulose coated lysine always were minimum. Compared with control group diet, total serum free amino acid absorption peak was delayed. Dissolution of lysine was prevented and the problem of out-sync of amino acid has been alleviated. There was a promotion on growth of Triploid crucian carp intaking diets adding hydroxymethyl cellulose coated lysine, and antioxidant capacity hasn’t been destroyed. The cost of Triploid crucian carp was been reduced.
