Rabies virus can caused rabies which could infect most mammal and lead toacute lethal encephalomyelitis. Worldwide at least55,000people died because ofrabies every year, and there were about2,000Chinese heath due to rabies virus everyyear. The main source of rabies virus infection in China is dogs, and there are a smallnumber of livestock and wild animals infected by rabies virus each year. Currently,there is no effective way to treat or prevent rabies, except vaccination. In order tounderstand the biological characteristics of the rabies virus this experiment measuredand analyzed the growth characteristics of12street rabies virus strains, and selected6stable proliferated street rabies virus strains. Based on the pathogencity differenceof these six strains, one strain with the highest titer was selected for mouse and doginfection experiment. The main contents include:1The culture and growth characteristics analysis of street rabies virus strains in ChinaCompare the growth characteristics difference of12street rabies virus strainsfrom different regions by measuring TCID50of different generations, continuouspassage through the cell culture and suckling mouse intracranial inoculation. Inwhich6strains’ viral titer increased stability and stabilized after20passages of cellculture or10generations of suckling mouse brain inoculation. HuNPB3strains havethe highest titer and stability as high as105.97TCID50/g or107.97TCID50/mL. RABV can bedetected in the third day cerebrum tissue, the sixth day cerebrum and cerebellumtissue, the ninth day cerebrum cerebellum and spinocerebellar tissue of intracranialinjection. As for10LD50hindlimb intramuscular, RABV can be detected in the thirdday spinocerebellar tissue, the sixth day spinocerebellar and cerebellum tissue, theninth day and the twelfth day cerebrum cerebellum and spinocerebellar tissue.Infected tissue has no obvious pathological changes.3The street rabies virus dog infection experimentUse dogs as experimental animal,106TCID50HuNPB3strains virus injected themasseter. Infected dogs showed typical clinical symptoms of paralytic rabies. Illnessbegan to show at the seventh day. Infected dog stopped eating and drinking at theeighth day and showed the symptoms of standing instability and depression.At thetenth day infected dog cannot raise its head, throat and tongue muscle contraction,vocal inconvenience. By the tenth day the dog forelimb paralysis and had difficultstanding. By the thirteenth day the infected dog was quadriplegic, then dead by thefifth day. RABV can be detected from the infected dog saliva seven days afterinfection. Necropsy the dead dog and RABV was detected in the brain, cerebellumand spinal cord, salivary glands, and corneal tissue, but not in heart, liver, spleen,lung and kidney tissue. Infected tissue has no obvious pathological changes...
