Recently, a growing of studies suggest nerve growth factor (NGF) can affect thereproductive activity. From this foundation, we study the effect of NGF onproliferation and secretion of progesterone in Kunming mice ovarian granulosacells.1Primary mouse ovarian granulosa cells were cultured successfully.Immunofluorescence was used to identify granulosa cells, MTS methods was used toportray the growth curve and detect the ability of cell proliferation.2Immunofluorescence was also used to observe the distributions of NGF andTrkA in granulosa cells.3MTS was used to analyze the different concentrations of NGF on proliferation ofmouse granulosa cells. The cells were treated with NGF of differentconcentrations(10,50,100and500ng/mL)for24h. The OD value of the cells wereidentified by micro-plate reader. Compared with the control group, each treatmentgroup promote the proliferation of granulosa cells with different degrees(p<0.05),the effect of (50ng/mL) NGF group on the granulosa cells proliferation was the mostsignificant(p<0.01).4In order to study the effect of NGF on secretion of progesterone, The granulosacells were treated with NGF, LH, K252, NGF+LH and NGF+K252for24h, then thecell culture media were collected to detect the progesterone concentration. Ourresults show that LH group increased progesterone concentrationssignificantly(p<0.01)compared with control. NGF group also increased theprogesterone concentration(p<0.05). RT-PCR was used to detect StAR andCYP11A1releative mRNA expression of LH group, the difference was significant (p<0.01). NGF group augmented StAR releative mRNA expression(p<0.05). It calsodecreased CYP11A1releative mRNA expression(p<0.01).In conclusion, The present study demonstrated that NGF can promoteproliferation of granulosa cells and can augment on sexul hormon secretion may byadjusting StAR mRNA expression.
