As a kind of quiescency stem cells, muscle satellite cells,existed in the betweencell membrane sheath and muscle fibres. As procursor cells, Muscle satellite cells fusewith muscle fiber and involve in injury response. Changes of muscle satellite cellimmune microenvironment directly relevant with treatment of autoimmune disease,and antiviral immunity with trichinella spiralis infection. The systemic knowledge ofsatellite cells immune microenvironment during trichinella spiralis infection isdeficient. In this study, we isolated the primary satellite cells, detected the cell byflow cytometry and fluorescence quantitative real-time PCR technology, furtheranalysis a series of changes in satellite cell immune microenvironment after infectedwith trichinella spiralis.Muscle satellite cells were isolated from BALB/c mice previously infected withtrichinella spiralis. High purity muscle satellite cells from hindlimb skeletal muscle ofinfected and non-infected mice were harvested by a combination methods of Percollcontinuous centrifugation and differential adhesion The isolated muscle satellite cellswere cultured and identified by Desmin immunocytochemical dyeing in vitro.Immunological character of the cells in4d,21d and35d were analysised by Real-time PCR and FCM. FCM analysis showed that B7-H1of muscle satellite cells wassignificant increased after trichinella spiralis infection, significantly reduced after35d of cyst formation and inflammation alleviation. The results of Real-time PCRevidenced that TLR3expression of muscle satellite cells was upregulated aftertrichinella spiralis infection. Its expression was apparently upregulated in the21dayof trichinella spiralis infection. The three subtypes of PPARs: PPAR α, PPAR β andPPAR γ expression also increased in different degree.This study shows that infection with trichinella spiralis can increase theexpression of TLR3, B7-H1, PPAR α, PPAR β and PPAR γand decreased expression of ICOSL. The immunology change can be concluded that infection with trichinellaspiralis enhance the ability of antiviral, anti-inflammatory and immune, strengthen theglycometabolism of satellite cells. The study will further improve the understandingon the biological characteristics in trichinella spiralis parasitic life,and provideexperimental evidence in immunology to treat autoimmune disease using trichinellaspiralis or product, and be offer a possible reference on study the antiviral immunitymechanism by trichinella spiralis infection. The results of the study will positivededicate to the application of trichinella spiralis or product in controlling musclesatellite cells as a biological agent and the pathogenesis study of muscle injury relatedto muscle satellite cell. The study also positive contribute to treatment of muscledysfunction, diabetes and so on by trichinella spiralis or product.
