Nutrition is one of the key factors affecting reproduction of insects. The nutrition regulation of vitellogenin (Vg) is particularly relevant to reproduction. By analyzing the structure of insect vitellogenin, the main factors affecting the insect growth can be identified, which may be used to pst control practices. The Vg of Nesidiocoris tenuis was cloned in this study, compared with other insects in phylogeny. By comparing the expression of Vg gene of Nesidiocoris tenuis feeding different diets with RT-PCR, it is found that the nutrition can affect the expression of Vg. In this thesis we obtain following results and conclusions.1The cloning of Vg gene of Nesidiocoris tenuisBy comparing Vg gene of different insects, the degenerate primers were designed for PCR, and the5’and3’Vg of Nesidiocoris tenuis was cloned with RACE. The length of the vitellogenin gene was6130bp, and of its open reading frame (ORF:57-5975)4422bp; it encoded1972amino acids. The former17amino acid residues was the signal peptide sequence. Removing the signal peptide, the predicted molecular weight of the protein was approximately216KD, and the isoelectric point of approximately9.12. Serine (S):15.5%, aspartic acid (N):5.22%, glutamine (Q):7.09%, alanine (A):9.83%.2Expression of Vg gene of Nesidiocoris tenuis feeding different dietsWith the method of qPCR, the expression of Vg gene of Nesidiocoris tenuis feeding Corcyra cephalonica (Stainton) egg, Antherea pernyi and yeast hydro lyat was compared. The result of feeding A was1.40904, feeding B0.00615and feeding C0.000225. Obveriously, the Vg expression by feeding A was higher than feeding B or C. Meanwhile, the Vg expression by feeding B was considerablely higher than feeding C. That is, the fecundity of Nesidiocoris tenuis feeding Corcyra cephalonica egg was more than feeding Antherea pernyi, and more than feeding yeast hydrolyat. This experiment suggested that Vg expression of Nesidiocoris tenuis feeding different diets can vary significantly, indicating direct effects of the Vg of Nesidiocoris tenuis on the reproduction.
