BackgroudImmune Thrombocytopenia is an autoimmune disease, the Immune function of patients islow.that previous, its pathogenesis is mainly B cells humoral Immune mediated. But in recentyears, more and more of the study show that, T cells play an important role in the immunemechanism of ITP in its morbidity and progress. According to whether the cells express CD4orCD8molecules, T cells can be divided into CD4~+T and CD8~+T cells. They differentiation intodifferent effect cells and play different immune effect. In normal circumstances, CD4~+/CD8~+constant proportion, and to maintain the normal cellular immune mechanism. Patients of ITPoften appear CD4~+/CD8~+ratio imbalance and functional disorder due to its dysimmunity.Thus, to analysis of the T cells subsets and the cytokines they secretion, can understand theimmune function condition of T cells. T cells (antigen) Receptor (TCR) is the characteristic signof all T cells.the role of TCR is to identify the antigen. According to the different with thepeptide chains, TCR can be divided into two types for TCRα β and TCR γ δ. Normally,thehuman body most express T cells for TCR α β calling α β T cells. All the T cells surface expressCD3molecules, TCR Combine with CD3molecules by means of a covalent bond to form theTCR-CD3compound. Both at home and abroad in recent years, using Genescan technology totest the length of CD3determine in T cells is an idea to test T cells cloned proliferation and tounderstand TCRV α β families express. this method is sensitive, accurate and objective.In order to understand the immune function condition of T cells in ITP patients, this paperdiscusses the effect of T cells in ITP about its occurrence, development and prognosis.Thisstudy analyzes the peripheral blood T cells subsets and the cytokines they differentiation in ITPpatients,and also the expression and cloning features in7cases of ITP patients before andafter treatment.The research content and methodsUsing Flow cytometric (FCM) to detect the T cell the subgroup and the cytokines theydifferentiation of peripheral blood in ITP and normal controls.Using statistics software SPSS toanalyze the data.To get the T cell receptor expression and cloning features before and aftertreatment in7patient of ITP With RT-PCR-Genescan technical.The results1.the immune function in patients with ITP is low, prompting T cells function disorder. For the33patients in this study,the results showed that the proportion of CD4~+T cells in patients withITP was lower than those in the healthy controls. The CD8~+T cells of ITP patients was somehowrised.It have statistically significant to normal control group. The CD3~+T cells and the CD4~+/CD8~+no statistical difference.TheIFN‐γ and IL-2were higher in ITP patients, and the ratioof them has no statistical difference.2.24cases of ITP patients, expressed0-25TCRV α family,and0-24TCRV β family. There areone or more cloning proliferation T cells with TVRVα OR TCRVβin24ITP patients, includingoligoclonal,oligoclonal trend and bioclonal patterns.The expression of the family and cloning in7patients with ITP are part of the improved after treatment. conclusion1.immune thrombocytopenia patients have immune function disorder, appear to T cellularimmune function defects. The level of T cells subgroup and the Cell factors they differentiationare changed.2.In ITP patients, The skew distribution and clonal expansion of TCR V a、V B subfamily T cellsis exist.part of the situation can improve after treatment.
