Objective:Through experimental study on the effect of tubulo interstitium injur of IgA nephropathy (IgAN) in rat with SHEN-Y AN-ZHI-XUE pill, further discusses this sickness to have the devel opment mechanism,for SHEN-YAN-ZHI-XUE pill toimprove tubuloin terstitialinjury mechanism to provide a more thorough theoretical an d experimental basis.Methods:Random will be divided into blank rats group, the model group, the control group (tripterygium wilfordii group), nephritis hemostatic pill is tall, medium, low dose group.Model:through the rats bovine serum albumin acidification oral water and tail intravenous fat polysaccharides and subcutaneous injection of carbon tetrachloride, castor oil compound method to establish IgAnephropathy disease model. Observation SHEN-YAN-ZHI-XUEpill to rat urine IgA nephropathy RBC count,24-hour urinary protein quantitative, kidney function, liver function andthe influence of the index, at the same time use light, immunefluorescence, immunohistochemical, RT-PCR and Motic image3.2image analysis software kidney tissues were observed (renal tubular-interstitial) pathology, mesangial area IgA deposition and a smooth muscle actin (a-SMA) and angiotensin receptorⅡ1(AT1), Ⅱ angiotensin receptor2(AT2) in renal tubular-mesenchymal expression and quantitative analysis.Results:SHEN-YAN-ZHI-XUE pill can reduce IgA nephropathy disease rat urine RBC count, reduce blood creatinine and urea nitro gen and tripterygium wilfordii group was significant difference (P<0.05), reduce albuminuria and tripterygium wilfordii group has no significant difference (P>0.05), and no tripterygium wilfordii muc h of the damage of liver function glucoside side effects; Can redu ce IgAnephropathy disease rat glomerular and renal tubular mesench ymal pathological damage; Canreducea-SMA、AT1and AT2IgAnep hropathy disease in rat renal tubular mesenchymal expression. One s hot high dose group nephritisbleeding curative effect is distinct, dos e group of in have apparent curative effect, small dose group of ef ficacy not sure.Conclusion:1.SHEN-YAN-ZHI-XUE pill can significantly reduc e IgA kidney disease rat models of red blood cells, to reduce the u rine serum creatinine and urea nitrogen, curative effect is better tha n tripterygium wilfordii control group, reduce albuminuria, and the control group tripterygium wilfordii treatment group, and has no tri pterygium wilfordii glucoside of liver function of more than the sid e effects of the damage.2.SHEN-YAN-ZHI-XUE pillcan relieve ren al tubular mesenchymal pathological damage.3. SHEN-YAN-ZHI-X UE pill can reduce the alpha SMA, and Ang II receptors in the Ig A kidney disease rat renal tubular mesenchymal expression.4. SHE N-YAN-ZHI-XUE pilll may adjust the regulation of alpha SMA, the activity of inflammation and reduce Ang II AT1AT2receptors an d the level, to reduce renal tubular mesenchymal damage.
