Anxiety symptoms such as insomnia constantly trouble modern peoples normal life, so research on the plants which have improving sleep functions is increasingly urgent. Semen Ziziphi Spinosae(SZS) is commonly used herb which could help healed insomnia and anxiety. SZS are the dried and riped seeds of Rharnnaceae herbs husjujuba Mill.var spionsa(Bunge)Hu. They can benefit the liver, calm the nervous, decrease the perspiration and increase the saliva. Their function was first recorded in "Shen Nong’s Herbal Classic". Modern pharmacology study show that they have excellent effects at the aspects of anti-arrhythmic action, improve microcirculation, strengthening organism immune system, etc. Jujuboside was always thought to be effective site with function above-mentioned. But according to researches in recent years, volatile oil also had the before-mentioned function such as sedation and hypnotism, antiatheroscloresis, anticonvulsant. So it must to be important that doing research on the extraction process of volatile oil, establish fingerprint, the preparation and development, quality control in compound preparation of Chinese tradition medicine.The studies in this thesis are summarized as follows:A rapid and simple method for the determination of volatile constituents in SZS was established by Head-space Solid-Phase Microextraction(HS-SPME)-GC-MS. HS-SPME-GC-MS conditions, including SPME fibre types, sample amount, temperature and time of extraction and desorption have been investigated and evaluated. The method is simple, fast and accurate, and it is suitable for the determination of the volatile constituents in SZS.The total oil from SZS was extracted by using supercritical carbon dioxide fluid(SCF) as extraction solvent, and the effects of the extraction parameters of pressure, temperature, time and entrainer amount were optimized according extraction yield. Meanwile, for discovering the variation of volatile constituents in all extaction from SZS, the volatile constituents were real-time tracked and analyzed by HS-SPME-GC-MS. The result indicated that the change of extraction condition not only influence yield of oil but also content of volatile compositions in oil. Among the extraction parameters, extraction pressure was the most affect on type and content of volatile constituents in oil from SZS. The method is simple, fast and good reproducibility. So it was suitable for the real-time tracking analysis of the volatile constituents in oil from SZS by supercritical carbon dioxide fluid extract (SCE), and lay a foundation for the industry-scale production of extracting oil from SZS by SCE. In addition the volatile constituents in oil from SZS by SCE have compared with the volatile constituents in SZS by HS-SPME directly and the volatile constituents in oil by steam distillation, the result showed that the variety of volatile constituents extracted was the most by HS-SPME directed from SZS.Volatile components in two compound preparation of SZS were extracted by HS-SPME optimized and analyzed by GC-MS, and compared with the results of original SZS by means of the same methords. Then the two compound preparation of SZS were extracted by SCE optimized. The yield of oil by SCE was compared with the results of original SZS by means of the same methords. The result indicated that volatile components of the two compound preparation of SZS with exactly the same ingredients were so different, and different from the volatile components of original SZS, in addition the yield of extract by SCE from the two compound preparation of SZS were far lower than the yield by means of the same methords from the original SZS. The results show that the loss of volatile oil were seriously during the preparation of the two compound preparation of SZS.
