Objective The present study was designed to detect the expression of EN2indifferent prostate cancer cells(LNCAP、PC3、DU145)and Normal Human ProstateEpithelial Cell Line(RWPE-1). Further, we detect the expression of EN2in humanprostate cancer tissues and benign prostate hyperplasia (BPH) tissues, and investigatethe correlation between EN2expression and Pathology classification. Our objective isto explore the correlation of EN2and prostate cancer, intend to find a new marker forearly diagnosis, prevention and treatment of prostate cancer (Pca).Method Firstly, immunofluorescence and Reverse Transcription-PolymeraseChain Reaction (RT-PCR) were performed to detect the expression of protein andEN2mRNA in different prostate cancer cells(LNCAP、PC3、DU145)and NormalHuman Prostate Epithelial Cell Line(RWPE-1). Next, immunohisochemistry andWestern blotting were performed to detect the EN2in51prostate cancer tissuesamples and57benign prostate hyperplasia tissue samples provided by The FirstAffiliated Hospital of JiNan University. Also, we evaluated the correlation betweenEN2expression and Pathology classification.Result1. The level of protein and EN2mRNA in prostate cancer cells weresignificantly higher compared with Normal Human Prostate Epithelial Cell Line.2. EN2expression in androgen-dependent prostate cancer cell(LNCAP) ishigher than that in androgen-independent prostate cancer cells(PC3、DU145).3. Prostate cancer tissues had shown high EN2expression, but low or hardlyin BPH tissues.4. EN2expression has no direct association to the pathological grade ofprostate cancer.Conclusions Taken together, the abnormal expression of EN2is correlated tothe occurrence and development of prostate carcinoma. It may be a promisingmolecular marker for the early clinical diagnosis、prevision、treatment of human prostate cancer.
