Objective: To investigate the expression of apoptosis signaling molecules’ genes andproteins affected by Ni-Cr,Co-Cr,CP Ti,Pd-base and Au-Pt metal-ceramic alloys inmouse lung fibroblasts cell (L929).Methods:(1)Ni-Cr, Co-Cr, CP Ti, Pd-base and Au-Pt metal-ceramic alloys were castfor one time to obtain sheet-shaped specimens. DMEM was used to infuse5differentkinds of ceramic alloy’ specimens for72h to get5different kinds of ceramic alloy’leaching liquor.(2) The mRNAs’ expression of Caspase-3、Caspase-8、Caspase-9、Cytochrome C、Bax,Bcl-2、Fas、Fasl in L929cells which were cultured by5differentkinds of ceramic alloy’ leaching liquor for48h were detected by Real-time qPCR.(3)The proteins’ expression of Caspase-3and Caspase-9in L929cells which werecultured by5different kinds of ceramic alloy’ leaching liquor for48h were detectedby Western Blot.Results:(1) The mRNA’s expression of Bcl-2was higher than negative control inAu-Pt group and Pd-base group (P<0.05). The mRNA’s expression of Bax was higherthan negative control in Ni-Cr group (P<0.05). The mRNA’s expression of Caspase-3and Caspase-9was higher than negative control in CP Ti group and Ni-Cr group(P<0.05). The mRNA’s expression of Caspase-8was lower than negative control in5different kinds of ceramic alloy groups (P<0.05). The mRNA’s expression ofCytochrome C was higher than negative control in5different kinds of ceramic alloygroups except Au-Pt group (P<0.05). The mRNA’s expression of Fas was higher thannegative control in5different kinds of ceramic alloy groups except CP Ti group(P<0.05). The mRNA’s expression of Fasl was higher than negative control in Ni-Crgroup, Pd-base group and Co-Cr group (P<0.05).(2) The protein’s expression ofCaspase-9was higher than negative control in5different kinds of ceramic alloygroups (P<0.05). The protein’s expression of Caspase-3was higher than negativecontrol in5different kinds of ceramic alloy groups except for Ni-Cr group (P<0.05).Conclusion: It is preliminarily estimated that:(1) different ions which were dissolved out from different kinds of ceramic alloy’ leaching liquor could induce apoptosissignaling molecules different expression in mRNA and protein;(2) the signal pathwayof apoptosis induced by ceramic alloys might not be extrinsic pathway;(3) the signalpathway of apoptosis induced by ceramic alloys might be intrinsic pathway;(4) thelower apoptosis rate induced by Au-Pt alloy and Pd-base alloy might relate to Bcl-2regulation.
