Objective:Analysis the metabolic differences among the thyroid papillary carcinoma, thyroidbenign tumor patients and healthy controls by nuclear magnetic resonance hydrogenspectrum; research the potential tumor metabolic markers and analysis its clinicalsignificance. It may provide a new way to diagnose the thyroid papillary carcinoma.Methods:1、According to the following inclusive criteria about tumour group, collecting theserum specimen of preoperative thyroid tumors including twenty cases thyroid papillarycarcinoma and twenty cases thyroid benign lesions. At the same period, collecting theserum specimen of20cases volunteers from medical examination center according to thefollowing inclusive criteria about control group. All the serum specimens were stored ineighty degree below zero.2、Analysis the serum specimen of above sixty patients with Switzerland Bruker500Hz NMR3、To phase and baseline correction use Topspin (2.1)software,to integral use AMIXsoftware (V3.9.11) between0.50-9.0ppm after calibration of the spectra,the width is0.005ppm.4、Analysis the differences about endogenous metabolites between the treatmentgroup and control group by PCA and PLS-DA with SIMCA-P+.Results:1、The data of thyroid papillary carcinoma and healthy control group have goodclustering in PCA. Discrimination model is established. By the PLS-DA cross validationand permutation test proving, the model is correct(p<0.05). Significant metabolites changes is judged by OPLS-DA analysis and Pearson’s product-moment correlationcoefficient.Compared with control group, the content of metabolites in the serum ofthyroid papillary carcinoma group are increased including valine, leucine, isoleucine,lactic acid, alanine, glutamic acid, lysine, glycine, but the lipids, choline, tyrosine arereduced.2、The data of thyroid benign tumor and healthy control group have good clusteringin PCA. Discrimination model is established. By the PLS-DA cross validation andpermutation test proving, the model is correct(p<0.05). Significant metabolites changes isjudged by OPLS-DA analysis and Pearson’s product-moment correlationcoefficient.Compared with control group, The content of metabolites in the serum ofthyroid benign tumor group are increased including Trimethyl glycine, tyrosine,phenylalanine, valine, leucine, isoleucine, lactic acid, alanine, glutamic acid, but theLipids and lysine are reduced.3、The data of thyroid papillary carcinoma and thyroid benign tumor group have nogood clustering in PCA. Discrimination model is established. By the PLS-DA crossvalidation and permutation test proving, the model is not correct(p>0.05).Conclusion:1、Compared with control group,there is an obvious metabolic difference in thyroidpapillary carcinoma group and thyroid benign tumor group.2、It is not only involve glucose metabolism but also the metabolism of lipid, aminoacids and nucleic acid.3、Metabonomics method can be applied to diagnose preoperative thyroid papillarycarcinoma based on nuclear magnetic resonance hydrogen spectrum and PCA/PLS–DA.4、It is not obvious about metabolic difference between thyroid benign tumor andthyroid papillary carcinoma with small sample analysis.
