Objective Through the detection of osteopontin (OPN) expression in degenerativespinal disease of vertebral cartilage endplate in the bone, The reason and the factors of changeof osteopontin in degenerative spinal disease of vertebral cartilage endplate degeneration, Inorder to elucidate the degenerative spinal disease of vertebral cartilage endplate bonedegeneration pathogenesis, pathological process and provide new clues. To investigate therelationship between OPN and the endplate MRI typing of Modic, to provide a newtheoretical basis for clinical diagnosis and treatment of degenerative spinal disease。Methods2011August to2012April, in Yinchuan city the first people’s HospitalDepartment of orthopedics diagnosis and operation treatment of10cases of spinal traumawithout cartilage endplate degeneration of bone specimens as control group. In13patientswith lumbar disc herniation, spinal stenosis, vertebral instability, endplate cartilage bonespecimens spondylolisthesis was experimental group. On the basis of MRI on bone signalModic lumbar endplate and endplate changes in patients with spinal degenerative disease,divided into three groups. Type1.I degeneration (edema):6cases on T1weighted imagenear the endplate region of vertebral endplate and low signal on T2weighted images, highsignal;2.Ⅱ degeneration (fat):5cases on T1weighted image and endplate area near endplatevertebral endplate increased relatively normal signal on T2weighted image, and is oftensignals are equal or slightly elevated bone marrow;3type Ⅲdegeneration (hardening) in2cases: in T1, T2weighted image and near the endplate region vertebral endplate signaldecreased. The resection specimens of each take about1grams of endplate bone, freezing inliquid nitrogen tank, to extract the total RNA, RT-PCRexpression of0PN mRNA determination;Image analysis was performed on the test results, the exposure of the imagescanning system, the band densities were analysis by gel imaging and analysis, using SPSS11.5statistical software for analysis of variance, significant differences in P<0.05.Results The normal group of edema group was significantly (P<0.05), differencebetween the normal group and fat group significantly (P<0.05), compared with normal groupand ossification group had significant difference (P<0.05), from normal to ossification group,the expression of OPN showed a rising trend, related that osteopontin and its characteristics.All the patients and normal controls were analyzed by T test, the results showed theexpression of OPN, the case group was significantly higher than that in normal group.Conclusions1.of osteopontin (OPN) expression in degenerative spinal disease ofvertebral cartilage endplate in the bone than spine trauma group without degenerativecartilage endplate in the high expression of bone.2.OPN and intervertebral disc endplate MRIexpression was positively correlated with Modic classification, play a role in the thickening,calcification and ossification of the cartilage endplate in.3.OPN can be used as a clinicaldiagnosis, therapeutic effect, changes of disease severity and the evaluation of degenerativespinal disease marker.
