Objective: To investigate the effects of visfatin on the expression offibronectin(FN) and typeⅠ collagen(COLⅠ) in rat mesangial cell line(HBZY-1) andits possible mechanism.Method: HBZY-1were cultured in vitro, then were incubated by differentconcentrations of visfatin or the same concentration at different time. The specificinhibitor of P38MAPK(SB203580) was also used. FN and COLⅠmRNA wereanalyzed by reverse transcriptase-PCR, and the expression of FN, COLⅠ andphosphorylated P38MAPK proteins were measured by western-blotting.Results:(1)The mRNA and proteins expression of FN all improved in cells afterHBZY-1were incubated by different concentrations of visfatin(P<0.05), from theconcentration of25ng/ml, the expression gradually increased with the increase ofconcentration of visfatin and the highest expression were at the concentration of100ng/ml;(2) FN mRNA and proteins were both up-regulated in cells after HBZY-1were incubated by the the100ng/ml visfatin at different time (P<0.05), from thetime of12h, the mRNA and proteins were further promoted with the prolongation oftime and the highest expression were at the time of24h;(3) The mRNA and proteinsexpression of COLⅠall improved in cells after HBZY-1were incubated by differentconcentrations of visfatin, the expression gradually increased with the increase ofconcentration of visfatin in the range of25-200ng/ml(P<0.05);(4)COLⅠ mRNA andproteins were both up-regulated in cells after HBZY-1were incubated by the100ng/ml visfatin at different time (P<0.05), the mRNA and proteins were furtherpromoted with the prolongation of time in the range of12-48h;(5) Visfatin upregulate the expression of phosphorylated P38MAPK protein(P<0.05), SB203580decreasedthe proteins expression of phosphorylated P38MAPK, FN and COLⅠ induced byvisfatin.Conclusion:(1)Visfatin increased the expression of FN and COLⅠin somedose-and-time dependent manner to promote glomerular fibrosis, involving in thepathogenesis of DN;(2) The role of visfatin above may be through the activation ofP38MAPK signal pathway.
