Methods42SPF grade healthy adult rats weredivided into3groups at random:6rats in control group(CON),18rats in surgerygroup(CLP),18rats in glutamine group(GLN),We made the septic mode via themeasure of cecal ligation and puncture (CLP).Then CLP and GLN were furtherdivided into3groups in3different time points:6h,12h and24h,each timepoint is consisted with6CLP rats and6GLN rats.6h later, collected serum andmyocardium in each group, detected the level of LDH(lactate dehydrogenase)ã€CK(Phosphocreatine kinase)ã€TNI(troponin I),Apoptosis index of myocardialcells was detected with TUNEL staining, the mRNA expression of genes p53andBcl-2were measured by polymerase chain reaction(PCR).Results Expression ofBcl-2in CLP group is lower than that in the CON group at all time point(P<0.05),expression of p53in CLP group is higher than that in the CON group atall time point (P<0.05). Expression of Bcl-2in GLN group is higher than that inthe CLP group at all time point (P<0.05),expression of p53in GLN group is lowerthan that in the CLP group at all time point (P<0.05).Apoptosis index ofmyocardial cells in CLP group is higher than that in the CON group at all timepoint (P<0.05),apoptosis index of myocardial cells in GLN group is lower thanthat in the CLP group at all time point (P<0.05). Compared with CON group, Thelevel of LDHã€CKã€TNI in serum in CLP group and GLN group were significantlyhigher(P<0.05); Compared with CLP group, the level of LDHã€CKã€TNI in serum in GLN group was significantly lower(P<0.05).Conclusion1.Glutamine cansignificantly reduce the level of LDHã€CKã€TNI in serum of septic rats, and haveprotective effect on myocardium.2. Myocardial apoptosis in septic rats wereincreased significantly, glutamine could inhibit the apoptosis of myocardial cell inseptic rats via affecting the expression of genes Bcl-2and p53. | | Keywords/Search Tags: | glutamine, septosis, LDH, CK, TNI, Bcl-2, p53 | | Related items |
