Objective To discuss the relationship between FSH and Wnt/β-catenin inEpithelial Ovarian Cancer (EOC) Cell Lines by examining whether FSH caninduce the proliferation of Epithelial Ovarian Cancer(EOC) Cell Lines SKOV3and HO8910and the expression of β-catenin in each of these ovarian cancer celllines stimulated by FSH.Methods After cultured SKOV3and HO8910being treated by FSH ofdifferent concentration (20,40,60,80,160mIu/ml) at different time(24,48,72h), the absorbency of the cells was detected by methyl thiazolyl tetrazolium(MTT). Taking FSH of the best concentration as experimental group andequivalent cell culture medium as control group to detect the expression ofβ-catenin in each of the cell lines effected by FSH by using immunofluorescence.To detect the expression of β-catenin with Western Blot by respectively takingFSH of different concentration (20,40,60,80mIu/ml) and equivalent cellculture medium cultivated for48hours as experimental group and control group.To conduct quantitative analysis by using Quantity-one to evaluate the value ofgray scale performed in protein band.Results FSH could induce the proliferation effects in ovarian cancer cellline SKOV3and HO8910. The proliferation of the HO8910cells stimulated byFSH of the concentrations of20,40,60mIu/ml for48hours was statisticallysignifican(t P<0.05).The proliferation of the SKOV3cells stimulated by FSH of theconcentration of40mIu/ml for48hours was the most statistically significant (P<0.05). Positive β-catenin expression was found in the nucleus and cytoplasmof both SKOV3and HO8910cell lines. After being stimulated by FSH for48hours, the cells of mean density with positive β-catenin expression increasedsignificantly(P<0.05)in SKOV3and HO8910. β-catenin expression was found inSKOV3and HO8910cell lines. The β-catenin expression of the HO8910cellsstimulated by the concentrations of20,40mIu/ml for48hours increasedsignificantly(P<0.05). The β-catenin expression of the SKOV3cells increasedsignificantly only after being stimulated by a concentration of20mIu/ml for48hours(P<0.05).Conclusions FSH could induce the proliferation effects in ovarian cancer cellline SKOV3and HO8910under certain circumstances. When the concentration ofa48-hour stimulation was40mIu/ml, the proliferation of cells was obvious. Wefound that the β-catenin expression was in the cytoplasmic and nuclear of SKOV3and HO8910. FSH could induce the increase of β-catenin expression in ovariancancer cell line SKOV3and HO8910.These results suggest that FSH induce theproliferation phenomena in EOC cell line through the Wnt signaling pathway.
