Objective:Present work aims to observe the expressions of TGF-B3、LN and RLX in front vaginal wall and uterosacral ligament with pelvic floor dysfunction (PFD), to detect biochemical changes in front vaginal wall and uterosacral ligament, and to explore whether the alteration in TGF-β3、LN and RLX in front vaginal wall tissue and uterosacral ligament tissue expression could contribute to pelvic floor dysfunction;Methods:Biopsy specimens of vaginal wall tissue and uterosacral ligament tissue expression were obtained from30patients with SUI,30patients with POP and15women undergoing without SUI and POP as control group. during operation vaginal wall and uterosacral ligament’s structure was examined with routine histological techniques. Reverse transcription polymerase chain reaction(RT—PCR) and western-blot to quantitatively determining the expression mRNA and protein level of TGF-β3、lamin and RelaxinResults:1. Real-time PCR(1) Compared with the control group, the expressions of lamin mRNA were lower and TGF-(33and Relaxin were higher in group POP and group SUI, this difference is of significance to statistics (p<0.001).(2) The expressions of TGF-β3、lamin and Relaxin in front vaginal wall and uterosacral ligament m-RNA in the group POP and group SUI has no statistical significance (P>0.05).2. Western blot detect(1) Compared with the control group, the expressions of lamin mRNA in these two groups had reduced and TGF-β3and Relaxin had raised both in front vaginal wall and uterosacral ligament, the difference is statistically significant (p<0.001).(2) Compared group POP with group SUI, there is no significance to statistics in terms of the expressions of LN in front vaginal wall (p=0.067),But in uterosacral ligament this difference is significance to statistics (p<0.001), group SUI is lower than group POP.(3) Compared the TGF-β3in the two groups, group SUI is higher than group POP both in front vaginal wall and uterosacral ligament, this difference is significance to statistics (p<0.001, p<0.001).(4)Compared the Relaxin in the two groups, group SUI is higher than group POP in front vaginal wall, but group SUI is lower than group POP in uterosacral ligament,this difference is significance to statistics(p<0.001, p<0.001).Conclusions:The occurrence of female pelvic floor functional disorder is related with the reduction of the expression of lamin and the heighten of Relaxin and TGF-β3in female vaginal wall and uterosacral ligament.
